Varietal Resistance to Stem Rot of Rice

Abstract/ Executive Summary:

The reaction of Sclerotium orvzee was tested on 25 mutant strains of Nlzersail along with Nizerseil mother as 8 check variety by cut stem (sheathed and unsheathed) inoculation method. The lesions on unsheathed cut stem were larger than on sheathed cut stem. Out of 26 tested materials. 17 materials were found to be resistant and 6 materials were moderately resistant in both the methods. Two materiels were graded to be moderately resistant and susceptible in sheathed cut stem and unsheathed cut stem inoculation methods respectively. Only a single material recorded to be resistant in sheathed cut stem and moderately resistant to unsheathed cut stem inoculation methods

Objectives:

To find out the degree of resistance among 25 mutant strains of Neizersail along with the mother ( cheek variety) against the disease.

Methodology:

Isolation of the pathogen: Before flowering of rice plants. the pathogen was isolated from the affected Ie af sheaths as described for sheath blight (Amin 1975). The mycelium is white to olive- in colour and the sclerotia are brown to black. Inoculum preparation: The stock culture was transfered to plates containing PDA medium and incubated at 25- 30⁰ C for 10 days. Inoculation test: The materials for the study comprised of 25 mutant strains originated through gamma ray irradiation of the variety Nizersail along with Nizersail mother as a check variety. The material was collected from the Breeding Division, Institute of Nuclear Agriculture, Mymensingh. One month old seedlings were transplanted in plots of randomized block design having three replications at the experimental farm of the Institute of Nuclear Agriculture, Mymensingh during Kharif season of 1979. Two seedlings per hill was transplanted at a spacing of 23 cm x 20.5 cm. Plants at flowering stage were tested by the following two methods: 1) Sheathed cut stem method, Ten cut stems each about 30 cm long were randomly collected from each block for each strain at the time of flowering and the internodes were wounded by needle punctures. A small block of ten days old fungal culture with sclerotia was placed on the wound site and wrapped by a piece of moist cotton. The entire cut stems were covered by a polyethylene bag and placed over a pan of shallow water to maintain high humidity and kept at room temperature. Records were noted ten days after inoculation.

This investigation considered the unsheathed cut stem inoculation method as the most efficient method for scoring the rice plants against stem rot disease.
