Allelic Relationship for Bacterial Blight Resistance in Rice

Abstract/ Executive Summary:

Inheritance of bacterial blight (BB) resistance caused by Xanihomonas campestris pv. oryzae in three resistant lines namely BRI61-2B-25, RP633-76-1 and BR319-1-HRll were studied. The test materials were crossed among themselves and with three susceptible local cultivars namely Dharial, Marichbati and Panbira in all possible combinations. Disease evaluation 2- weeks after inoculation of the parents, F_I, F₂ and F₃ generations indicated that the resistance in the three lines was controlled by a single recessive gene. The gene for BB resistance in the three resistant lines was allelic and no influence of the cytoplasmic factor was evident.

Objectives:

To investigate genetics of resistance, allelic relationship of the genes and the influence of maternal factors, if any, on resistance to BB in three resistant breeding lines of rice.

Methodology:

This study was conducted at Bangladesh Rice Research Institute farm, Gazipur during 1983-1985. Three resistant lines namely BRI61-2B-2S, RP633-76-1 and BR319-1-HRll and three susceptible local varieties namely Dharial, Marichbati and Panbira were used. Crosses were made in October, 1983, and June, 1984 in all possible combinations except among the susceptible varieties. Fi'S were confirmed in November, 1985 by growing and comparing with the respective parents, F₂'S and F₃'S were grown in February-June and July- November, 1985, respectively. Fertilizers at 120:60:40 kg NPK/ha. N, P and K were used in the form of urea, triple super phosphate and muriate of potash, respectively. Urea was applied at three equal installments. The first at 15 days after transplanting. The second at 35 days after transplanting and the last at panicle initiation stage. All other fertilizers were applied at the time of final land preparation. Test population sizes were 10 plants for each parent and F₁,230-524 plants for each F₂ and 60-90 lines for each F₃ family rows. For easy comparison of disease evaluation TNI and RP633-76-1 were included as standard susceptible and resistant checks respectively. The test materials were inoculated following clipping method (Kauffman et al, 1973) at panicle initiation to booting stage with 2-day old culture of BX05. It is a virulent bacterial isolate predominant in north-western part of Bangladesh and was obtained from the plant pathology division of BRRI. Disease severity was evaluated two weeks after inoculation. Individual plant was scored visually following the standard evaluation system for rice (IRRI, 1980).
