Selenium Level in Various Tissues of Guinea-pigs in Experimental Selenium Toxicity

Abstract/ Executive Summary:

The experiment was conducted on 131 recently weaned guinea-pigs. Organic selenosis was induced by feeding on water-soaked selenium enriched barley (containing 6.10 ppm selenium); inorganic selenosis by feeding ordinary barley (containing 0.86 ppm selenium) to which sodium selenite (@ 30 ppm) was added; the treated group was kept on ordinary barley to which sodium selenite (@ 30 porn) and sodium arsenite (@ 10 ppm) were added. Three randomly selected animals from each group were killed at 20 days intervar and selenium level of liver, kidney. spleen and hair of these animals were estimated. A well marked elevation of selenium concentration was noticed in the examined tissues in all the three experimental groups with highest values in inorganic group followed by organic and sodium arsenite treated groups. A continuous increase in selenium level of hair was noticed throughout the experiment in all groups. Tha selenlurn level in the kidney was alwava found higher than that of liver in both the toxicoses.

Objectives:

The Selenium levels in liver, kidney. spleen and hair were estimated' in the experimentally induced organic and inorganic Se toxicity in guinea-pigs, as well as, in sodium arsenite treated inorganic Se toxicity.

Methodology:

The experiment was conducted on 131 recently weaned guinea-pigs (albino-English breed). At 0 day. i. e. prior to the actual experimentation three randomly selected animals were killed and the liver. kidney. spleen and hair were collected for Se estimation. The remaining 128 guinea-pigs were divided randomly into forur groups. Organic Se toxicity was induced in the guinea-pigs of group A (40) by feeding on 48 hrs, Water-soaked selenium enriched barley (containing 5.10 ppm Se) to which vitamin C ( @ 0.2% of dry feed) was added throughout the 60 days course of the experiment. Animals in group B (40) were fed on water-soaked ordinary barley (containing 0.86 ppm Se) to Which sodium selenite (@ 30 ppm of dry feed) and vitamin C (@ 0.2% of dry feed) was added for first 23 days. after which. the sodium selenite was reduced to 15 ppm and continued till the end of the experiment. The guinea-pigs of group C ( 24) were kept on water soaked ordinary barley with vitamin C and served as control. Group D animals (24) were fed on water-soaked ordinary barley mixed with sodium selenite (@30 ppm of dry feed). vitamin C (@ 02% of dry feed) and and sodium arsenite (@10 ppm of dry feed) throughout the experimental period. During the course of the experiment a total of 22. 22 and 9 animals died from group A. B. and 0 respectively and the tissues of these animals have been excluded from this estimation. Three randomly selected animals from each groups were killed at 10 days interval and the tissues like liver,kidney spleen and hair were subjected to Se estimation only from those collected at 20th, 40th and 60th day following the method of Cummins et al. (1965). and Levesque and Vendette (1971) by using Spsctronic 20. The calibration curves for different concentrations of Se were used to determine the values of unknown samples.

Achievement/Findings:

Although the sodium selenite concentration in the feed supplied to the animals of group 0 was higher than that of group B but the Se level in various tissues of the treated group (D) was relatively lower. This indicated that the sodium arsenite had partial protective effect against inorganic selenosis.
