Determination of Oestrogenic Activity in Durbagrass (cynodon Dactylon)

Abstract/ Executive Summary:

Durba grass is comonly fed to the milch cow with the idea to increase milk yield. The oestrogenic principles were extracted from this grass and detected by 'Frog Test'. The oestrogenic activity of durba extract was standardized on 42 virgin female rats, noting the increased uterine weight and comparing against the response produced by known doses of diethylstilboestrol. The average uterine weight of control groups i. e. no treatment & treatment with olive oil were 10.1 mg. and 13.5 mg. The average uterine weight gain to different doses of diethylstilboestrol (0.02 mg. to 0.05 mg.) was 33.8 mg., 40.6 mg., 51.5 mg and 68.3 mg. respectively. The average response produced by 3gm equivalent of durba extract was 24.3mg which was lesser than the response produced by tbe minimum dose of diethylstiboestrol (33.8mg.) but higher than those in the control groups (10.1 mg. and 13.5 mg.) Thus, the oestrogenic activity found in durba extract might have relationship with the increased milk yield in cows fed on durba grass.

Objectives:

To determine whether increase in milk yield in lactating cows was due to feeding durba grass (Cynodon dactyIon) contairung oestrogenic activity.

Methodology:

Collection and preparation of sample: The succulent durba grass was collected during the rainy season from the lawns of the University Campus. It was cleaned, washed with water, sun dried, oven dried at 45°C and pulvurized to 40 mesh powder. Diethylstilboestrol was procured from the Bangladesh Pharmaceutical Industry, Tongi, Dacca. The method applied in determining the oestrogenic activity in durba grass consisted of 3 steps. 1. Extraction of oestrogenic principles from durba grass. 2. Detection of oestrogenic principles: Presence of oestrogenic activity in the extracts were detected by 'Frog Test' by studying the spermatogenic response in male frogs comparing against the dieth- ylstilboestrol. Oestrogenic extracts from durba grass were injected into the dorsal lymph sac of one group of male frogs. Dietbylstilboestrol was also injected into the 2nd group of male frogs. Spermatogenic response produced by durba extracts was similar to that of diethylstilboestrol indicating the presennce of oestrogenic principles in the sample. 3. Bio-assay of oestrogenic activity found in Durba extract: Oestrogenic activity of the extracts was standardized on about 20 days old virgin female rats noting the weight gain of uteri and comparing against the response produced by known doses of injections of diethyistilbocstroi. During the experimental period rats were maintained on a ration consisting of wheat bran, gram, fish meal and common salt. Forty two rats were used in this experiment and were divided into 7 groups consisting of 6 rats each. Group 1 and 2 were used as control (Group 1 was not treated with and group 2 was treated with Olive Oil). Group 3 to 6 received injections of diethylstilboestrol diluted in olive oil in doses of 0.02 microgram to 0.05 microgram per rat per day. Group No.7 was treated with 3gm equivalent of durba extracts also diluted in olive oil. Injections were given subcutaneousIy over a period of 4 days only. Treated rats were killed 24 hours after the last Injections. Immediately the uteri were dissected out, cleaned, freed from the adhering tissues and dried pressing against filter papers and weighed.

The average response produced by 3 gm equivalent of durba extract was 24.3 mg which was lesser than the response produced by the minimum dose of diethylstiboestrol (33.8 mg.) but higher than those in the control groups (10.1 mg. and 13.5 mg.)
