Diallel Analysis of Oil and Protein Content of Brassica Campestris L. and B. Juncea L.

Abstract/ Executive Summary:

The nature and magnitude of genetic variances and cornbining ability for oil and protein content were studied in 6.parent diallel cross of B. campestris L. and B. Juncea L. The analysis of diallel table, graphical analysis, components of genetic variation and combining ability, all suggested the greater importance of additive variance for protein content of both species; but for oil content. both additive and non-additive genetic variances appeared to be equally important. The non-allelic gene inter- actions of a complementary type were detected for oil content. On average, over dominance and partial dominance were obtained for oil and protein content respectively. Only one effective factor was detected to condition each of the two characters. The best general combiners for oil and protein content of B. campestris L. were Tori-Iu and Pusa Kalyni ; and of B. juncea L. were Varuna and Rai-Monipuri respectively. One and 6 crosses respectively for protein and oil content of B. campestris L., 5 crosses for oil content of B. juncea L. showed significant positive sea effects. In B. carnpestris L., heritability was estimated to be 44 % and 45% for oil and protein content respectively. In B. juncea L., on the other hand, the corresponding values were estimated to be 50% and 76%.

Objectives:

In the present investigation, crosses between six varieties of B. campestris L. and between six varieties of B. juncea L. were made and these were studied in F₁ generation.

Methodology:

In two separate experiments, seeds of six varieties of B. campestrfs L. viz., Assam Local, Pusa Kalyni, Saphala, T.9, Tori-10, Tori-7 and their possible F I hybrids (without reciprocals) ; and seeds of six varieties of B. juncea L., viz., Varuna, KB.2, Laha-lOl, Rai-7, Rai-5, Rai- Monipuri and their possible F, hybrids ( without reciprocals), were grown at the experimental garden of Rajshahi Universitry, during 1974-75. The experimental design and layout were the same for both experiments. Each experiment had two replications, and in each replication there was a single plot of ten spaced plants per population. The distance between the plants and that between the plots was 30 em. Fully matured fruits from ten plants of each plot were collected and were bulked. After proper drying the seeds were put in paper bags and stored in desiccators. Duplicate samples of these seeds were used to deter- mine the percentages of oil and protein content. Oil was extracted by the solvent extraction method using a soxhlet extractor and petroleum ether (b. p. 40.60°C ) as solvent. From the defatted meal, the nitrogen content was determined by the microkjeldahl procedure. The percentage of crude protein was estimated by multi- plying the total percentage of nitrogen in the defatted meal by the factor 6.25. The data thus obtained for the oil and protein content were analysed by using the diallel techniques.
