Phytoplanktons of Pond Soils of Bangladesh Agricultural University Campus, Mymensingh

Abstract/ Executive Summary:

A preliminary survey was undertaken in order to isolate and identify phytoplankrons from pond soils of Bangladesh Agricultural University campus, Mymensingh. Soil samples were collected from four different cultivable perennial ponds located in different places of Bangladesh Agricultural University campus, Myrnensingh for study. Isolation of phytoplanktons were made following Chu's technique and identification was done under direct microscopic examination considering their morphological characters. Twelve different genera of phytoplanktons of which seven belonging to Cyanophyceae, three belonging to Chlorophyceae and two belonging to Xanthophyceae were identified. Among the identified genera Anabaena was found dominant in Cyanophyceae, Chlorella was found dominant in Chlorophyceae and Vaucheria was dominant in Xanthophyceae. Most probable number of phytoplanktons per gram of soil was also estimated which ranged from 1880 to 15300.

Objectives:

To determine the total phytoplankton population per gram of soiI.

Methodology:

Soil samples representing 0-15 cm depth and differing in pH, mechanical and chemical composition from four well distributed ponds were collected with the help of Ekman dredge. The ponds were designated arbitrarily as pond I, 2, 3 and 4 for convenience of presentation. After collection, the samples were divided into two partisan part for physical and mechanical analysis and other part for microbiological studies. The methods adopted for physical and mechanical analysis were as follows: Mechanical analysis of soil was done by pipette sampling method and the textural class was determined following Marshall's "Triangular coordinates" using international system. Soil pH' was measured with the help of glass electrode pH meter, the soil water ratio being maintained at 1 : 2.5 as described. Organic carbon in soil was estimated by wet oxidation method devised. Organic matter content was calculated by multiplying the percentage of organic carbon with the conventional factor 1.72. The isolation and identification of the phytoplankton was the major task of this study. The most probable number method was used for the estimation of the most probable number of phytoplankton per gram of soil. Dilution series of soil up to 10-5 were prepared and inoculated in the test tubes (2 for each dilution) containing Chu's medium. The inoculated test tubes were incubated under electric light shade of 10 sixty Watt bulbs maintaining: cm distances from each and 20 cm distances above the sample. After 30 days the algal growth was seen, picked up and examined under microscope. Most probable number of phytoplanktons per gram of soil was determined by using the Bristol's medium following probability table for determination of abundance of organisms in soil.

Achievement/Findings:

The value associated with the proper code, and located under the column designated V, is multiplied by the reciprocal of the center dilution to obtain the most probable number of phytoplanktons present per gram of soil. The number of phytoplanktons was calculated from the different types of soil which ranged from 1880 to 15300. The four ponds are suitable for fish culture but to draw concrete decision further studies should be needed.
