Effects of Some Antibiotics on the Grqwth of Fresh- Water Phytoplanktons and Their Use in Obtaining Pure Cultures of These Organisms.

Abstract/ Executive Summary:

The effect of three antibiotics, namely, Penicillin, Strepto- mycine and Kanamycin has been studied on the growth of ten freshwater phytoplanktons. A method has been devised to culture these algal species in bacteria free condition. Results have shown that Chlorella vulgaris and Ankistrodesmus falcatus are both extremely susceptible to even low doses of the antibiotics in inorganic culture media, whereas, the remaining eight phytoplanktons are comparatively less susceptible to these antibiotics in similar situation. But they all tolerated higher doses of the antibiotics in organic media. The experiment, therefore, proved that the toxicity of antibiotics decreases significantly in organic media as compared to that of inorganic media. Further, it was noted that application of antibiotics breaks-up coenobium of Scenedesmus quadricauda and this coenobium develops normally in absence of antibiotics.

Methodology:

Ten species of algae as listed below were collected from Lake Sagami, Japan and were isolated in unialgal cultures :_ 3. Chlamydomonas reinhardi 4. Pandorina morum 5. Chlorel1a vulgaris 6. Chlorella pyrenoidosa 7. Pediastrum boryanum 8. Scenedesmus quadricauda 9. Synedra ulna 10. Nitzschia palea 11. Microcystis aeruglnosn 12. Ankistrodesmus falcatus Two types of media were employed in the experiment, one was completely inorganic in composition (of Ichimura, 1967), the other coutain-d two org mic substances. namely, yeast extract, 15 mgj l and peptone 25 mg/l along with all the inorganic substances used in the inorganic medium. The' medium of Ichirnura (1967) allowed good growth of all species excepting Microcystis aer- uglnosa. To grow the Jatter successfully the medium of Ichimura (1967) was modified and Was employed in the ex- periments with this modification related to an addition of some micronutrients in the form of inner complex salts of ethylenediamine-tetracetic acid (EDT A), as suggested by Thomas and Keauss (1955). Hundred milligram of the stock solution of the complex contained 0.385 g. NaFe ; 0.071 g. EDTA. NaMn ; ().077 g. EOTA. Nl,CO; and 0.067 g. EOTA. Na!Zn. IO ml. of this stock solution was added to one litre of the culture medium. This yielded a concentration of 5 opm. of iron and I ppm. of the oth rr micronutrients in the medium. The pH was adjusted to 7.5 with KOH (If HN03.
