A Study on the Preservation of Bovine Semen at Room Temperatures

Abstract/ Executive Summary:

The semen for storage at room temperatures in different diluters and at 5°c in egg yolk citrate extender was collected by artificial vagina. Semen was diluted at the ratio of 1:20 in different cultures; corked and stored in dark environment. The method used was the daily removal of supernatant fluid at 24 hours interval from the storage tubes with addition of fresh fluids. pH of the diluted semen was measured before removal of supernatant fluid with nitrazine paper readily (pH hat paper) 86. The performance in cocoanut milk exteder and cocoanut milk egg yolk extender was poor. The factors like lacking of catalase, rapid fluctolysis because of high room temperature leading to increase accumulation of acid which inhibited enzyme action, denaturation of some and yet constituents of the diluents possible H₂0₂formation other factors were held responsible for unsatisfactory result with cocoanut milk extender and cocoanut milk egg yolk extender and these factors were less pronounced in case of other extenders. The pH and motlity percent were positively correlated while room temperatnre and motility percent were negatively correlated. Extenders Glucose citrate and Cornel University -16 were good upto 24 and 48 hours respectively.

Objectives:

To study the relative merits of the diluents like coconut milk-Na-citrate, coconut milk- egg-yolk, glucose-citrate-Nal+Ctrj-Kl and Cornell University-16 at room temperatures against the egg yolk citrate at refrigeration temperature (5⁰C) in maintaining bovine sperm motility.

Methodology:

Semen was collected three times a week from the stud bulls at the Artificial Insemination, Laboratory by Artificial Vagina method and in total 65 ejaculate were collected during the experimental period. Semen was evaluated immediately after collection by gross examination of ejaculates by observing the colour, consistency, swirl and wave motion. An average of 84% was estimated under 40X objective, The pH was 6.70 and Haemocytometeli was used for counting the sperm cells and the average sperm-count per o.m.m. was 16 x 10⁴ . Precautions were taken against cold shoe and high temperature. Each ml. of the ejaculates was split at room temperatures into equal portion and each portion was pipetted to 5 ml of the respective diluents in the ratio of 1 :20 and each ml. ejaculate contained 1352.4 million motile sperm cells and each ml. of the extenders after dilution contained on average 67 million motile sperms initially. An equal portion of semen was extended as control in Yolk-citrate diluent and preserved at 5°c. The data was analysed and the analysis of variance was done by using completely randomized design. Duncan's New Multiple Range Test was used to compare the treatment means. The correlation coefficient between percentages of motile spermatozoa and corresponding pH level within a specific range and that between perservation temperature and percent motility of spermatozoa after 24 hours of preservation were calculated.
