Selection Of, Mustard Plants ( Brassica Juncea L. ) with Low Content of Linolenic Acid.

Abstract/ Executive Summary:

Single plant selections for low linolenic acid content of mustard ( Brassica juncea L.) oil were made in the F₂. generation of the cross Rai--5x Rai--3. The range of variations for fatty acids were considerable. Two seeds were identified containing as low as 4.27 and 4.33 percent linolenic acid. Either oleic or linoleic (or both) has replaced linolenic acid content as a major constituent.

Methodology:

In this experiment, two mustard cultivars (Brassica juncea L.), Rai (R5) and Rai 3(R3), were used. These R5 and R3 parent plants, developed through pure line selections were originally taken from two different populations of mustard plants from Bangladesh. Normally, mustard takes 4-5 percent cross pollination. Therefore, one can conclude that cross-parents could show some degree of heterozygosity. The F ₁ seeds derived from the cross R5 x R3 were grown in I5 cm. diameter pots in the greenhouse starting from July, 22, 1976. An average temperature of 21°C was maintained throughout the growing period. At flowering stage, 60 x 20 cm diameter plastic bags were used to Cover the plants to ensure self pollination. Morphological characters of the population were noted for better selection practice. On November 22, 1976, the mustard seeds as F₂ were collected from the individual plants which Were analyzed by G LPC using the 'Half seed technique'. All seeds form 2 plants and only 10 seeds from each of selected 89 plants Were analyzed. The analysis for fatty acid content was done on the outer cotyledon by allowing the remainder of the embryo to develop into a normal plant for further work.

Achievement/Findings:

The comparison of results of selected and unselected cotyledons showed that either oleic or linoleic acid (or both) has replaced linolenic acid content as a major constituent .The rate of replacement was weak in some cases while strong in others, indicating that it will be possible to isolate seeds which are of high linolenic acid content and at the same time practically free from linolenic acid. However, the levels of other fatty acids showed no deviation. It is yet to be established whether this change in the content of linolenic acid was accompanied by any significant change in the oil content of the seed. However, this will be done in future. The present investigation disclosed the fact that further selection in the desired direction will give promise of success.
