Effect of Ethyl Methanesulphonate (ems )and Diethyl Sulfate ( Des) on Some Morphological Characters of Three Mustard Varieties in Ml Generation-i

Abstract/ Executive Summary:

Two separate experiments were conducted with EMS and dES during winter season of 1975 with a view to studying the effects of these chemicals on some of the characters of three mustared varieties. The results showed quite a high level of influence of these chamicals on various characteristics studied. All the three varieties under two experimental conditions showed negative response to the doses of EMS and dES for seedling emergence and plant height. In cases of days required for flowering and days required for maturity, positive responses of the varieties to both types of mutagens were observed. It was further interesting to note that the seedling emergence was more drastically influenced by EMS than by dES. Similar was the condition in days to flowering and days to maturity. In case of plant height the sensitivity of the varieties was almost constant in experiment EMS, but it was not so in dES experiment, In this case it was noted that two varieties ( V₁ and V₃ ) did not respond significantly to the doses of dES.

Objectives:

To study the effects of chemicals on some of the characters of three mustared varieties

Methodology:

The experiment was conducted at the Bangladesh Agricultural University Farm in rabi season, during the period from November, 1975 to March, 1976. The soil structure of the experimental plot was sandy loam and pH of the soil ranged from 6.5 to 6.6. Two chemicals namely, Ethyl metha- nesulphonate (EMS) and diethyl sulfate (dES) were used. Three Varieties of mustard of the species (Brassica juncea) namely, BAU.M/14,V1), BAU.M/19(Vw) and BAU.M/9(V s) were chosen as the experimental materials. Two separate experiments were con- ducted keeping the varieties common, where all conditions were same for the experiments except the levels of chemo- mutagens. 18 hours pre soaked seeds (Savin, et al. 1968) of three varieties were used. In the first experiment, the doses of EMS were 0 ; 0.16, 0.32,0.48 and 0•64 % ; and in the second experiment the same for dES Were 0, 0.16, 0.32, 0.48, 0.64 and 0.80%. In case of EMS, treatment of seeds with 0.807'~ resulted in no germination of seeds whereas dES showed sufficient germination at the same dose. One sample in each experiment was used as control. Thus, for the first and second experiment there were 5 and 6 treatments respectively. The treated seeds were thoroughly washed in running tap water before sowing in the field. Split-plot randomized block design with three replications was used for both experiments. The varieties Were placed . in the main plot and the doses of chemicals along with control were placed in the sub-plots, The unit plot size was 4.87 x 1.82 meters. The row to row and plant to plant distances were 30 em and 23 cm, respectively.
