Measurement of the Output of Spermatozoa in Urine of Rams

Abstract/ Executive Summary:

The daily output of spermatozoa was measured in the urine of two rams for 12 consecutive days. Single representative sample from the daily collection of urine was found to be sufficient for counting the number of spermatozoa without much error. A large between day variation in daily output of spermatozoa was observed in each ram along with a large variation in the concentration of spermatozoa in different urination within each day. It is realized that spermatozoa comes out through Spontaneous emissions of semen in the normal urine of ram; masturbation and ejaculation of semen by the ram during collection of urine may not contribute a major output of spermatozoa in the urine. The daily output of spermatozoa of rams can be measured from number of spermatozoa voided in the daily urine.

Objectives:

The present study was carried out to develop a suitable collection method and sampling technique for counting the number of spermatozoa voided in the urine. The variation in the daily output of spermatozoa and the probable masturbation and ejaculation of semen during the collection of urine were also investigated

Methodology:

A two year old Merino ram and a one year old Finnish Land race ram were used in this experiment. Both the rams had undergone a libido test prior to the start of the experiment. They were in good health and exhibited an ardent desire for mating. Ram, were fed on a pelleted complete ruminant diet, during the experimental period. For collection of urine, each ram was placed in metabolism crate, especially designed for sheep where the animal could stay quite comfortably about 3 feet above ground level for several days. The urine was collected by a water- proof flexible cone connected with a rubber tube. Five independent samples of 20 ml each Were taken from each day's collection of urine. The bottle containing daily collection of urine and saponin solution was shaken well before taking each sample. The total number of spermatozoa in daily urine was determined by haemocytometer count under 450 magnification

Achievement/Findings:

The rams were constantly observed during twenty four hours for a single day. A sample of about 10ml of urine was taken away during each urination and the remainder was allowed to pass through the cone to the bottle. A fresh cone was used after every urination and the prepuce washed and dried every time before placing the cone. The sample collected during urination and the whole urine in the bottle was examined under the microscope. It was noted that the Merino ram erected its penis three times at night but no sign of masturbation or any secretion of semen followed. The Finn ram showed no erection of penis in that night. Examinations on the sample of urine indicated that the spermatozoa were present in the urine of both rams but the concentration of spermatozoa in different' urination varied very much. It is realized that the rams may not masturbate against the cone fastened under the prepuce but the rate at which spermatozoa comes out in the normal urine differs very much between urination and follows irregular interval
