Effect of Inoculation of Mycorrhizal Fungi on the Growth and Survival of Pinus Oocarpa in the Nursery

Abstract/ Executive Summary:

Investigations were made to study the effect of inoculation of mycorrhizal fungi in the potted seedlings of Pinus oocarpa. The inoculation was carried out by spreading mycorrhizal soil collected from existing pine stand and by spreading liquid Hagem nutrient containing a mycorrhizal fungus grown in the same medium. The results indicated that the growth rate was significantly higher (P > 0.05) in the seedling spread with mycorrhizal soil. However, the survival of the seedlings though apparently higher in the same treatment, was not significant.

Objectives:

a) To determine the suitability of inoculation techniques for the establishment of mycorrhizal association.

b) To the impact of mycorrhizal association on the survival and growth responses of pine seedlings.

Methodology:

Studies were made with P. oocarpa as seeds of any other pines were not available. The seedlings were inoculated with mycorrhizal fungi by two methods. Observations in respect of height growth and mortality of the seedlings were recorded, at monthly intervals for six months. Heights of the seedlings were measured from cotyledonary scars to shoot apices. Natural top soil, free from debris was collected and sterilized at 0.16 KSC for one and cooled for filling up earthen pots (23 cm dia., 20 cm ht.), fumigated with formaldehyde, before setting the experiment. Six seeds were sown directly in the pots. The seeds were surface sterilized in alcohol bath before sowing. One vigorous seedling was retained in each pot after third week of germination and the rest were removed. The potted seedlings were placed in a green house under natural condition during the experimental period and were watered once a day. A distance of I m was maintained between the seedlings of different treatments. Inoculations were done after 4 weeks of germination. The soil inoculum was collected from the floor of a successfully established pine stand, planted ill 1970 in the South East aspect of the northern hill at FRI. Only the top 15cm soil was collected for use as inoculum in T₁. 125 g of these soil inoculum was added as top dressing to the sterilized soil of each pot containing seedlings. For T₂ corraloid roots of P. oocarpa seedlings raised in polyethylene bags 10 the nursery (inoculum source was the mycorrhizal soil of existing local pine plantation) of the SRD were collected for isolation of mycorrizae. The roots were then washed thoroughly to clean adhering debris and then surface sterilized in H₂O₂ solution. The roottips were then plated in Hagem's Nutrient Agar Medium (HAM) in order to obtain mycelial growth and incubated in about 23°C. The isolated fungus, was then cultured in liquid HAM for IS days at room temperature (27°C±5). Subsequently, 5 ml of this liquid culture was used directly as inoculum in each pot containing seedlings.

Achievement/Findings:

In the present study the soil containing mycorrhizal short roots was found to be effective as inoculum. It appears that liquid HAM inoculum did not succeed to establish proper symbiotic association.
