Radiosensitivity of Soybean (glysine Max L; Merrill - Ii. Effect of Gamma-rays on Varieties Bragg and Lee-74

Abstract/ Executive Summary:

The radiosensitivity of two Soybean varieties, Bragg and Lee-74 was measured in the Ml generation in terms of the reduction of seedling height, plant height, survival at maturity, pollen fertility, frequency of fertile plants and number of fruits per plant. All of the characters showed negative correlations with the gamma-ray doses. LD50 and RD50 values for all characters excepting seedling height were higher in Bragg than in Lee-74, indicating that Bragg is somewhat more radiosensitive than Lee-74. However, from the available data it is suggested that gamma-ray doses ranging between 20 and 30 kR would be optimal for both varieties in a mutation breeding programme.

Objectives:

To assess the radiosensitivity of soybean varieties; Bragg and Lee-74 and dose of gamma-rays for mutation breeding.

Methodology:

Air-dry seeds of Bragg and Lee-74 with about 12 % moisture content were subjected to 15, 20, 25, 30 and 35 kR doses of gamma-rays from a Co 60 source at Atomic Energy Centre, Dacca, keeping time coantnst. Both control and irradiated seeds were sown in the field. The layout was a randomized block design with five replication for each treatment and eight seeds for each replication. The usual cultural operations were carried out throughout the season to facilitate normal growth and development. To measure radiosensitivity, observations were recorded on seedling height, plant height, survival at maturity, pollen fertility, frequency of fertile plants and number of fruits per plant. Seedling height Was noted on the 22nd day from the date of sowing. Seedling height and plant height refer to the distance between the shoot apex and soil surface. The fertility of pollen grains was measured by using 2% acetocarmine as a staining solution following Lapins (1975). The fertility percentage was estimated for each plant in each dose using at least three thousand pollen grains and the mean of all the plants in a dose was calculated. The plants which were either fruitless or with seedless fruit were considered as sterile plants and those with fertile fruits were considered as fertile plants. The frequency of fertile plants was expressed as a percentage. Statistical analysis of the data was' done to find the correlation coefficients and regression equations. The expected LD/I~ (dose at which 50% lethality occurs) was estimated from the survival data and expected RD /I 0 (dose giving 50% reduction) was calculated from the data of other characters. "t" tests were made to determine the significance of differences between the correlation coefficients within the observed data.
