Experimental Production of Duck Plague Vaccine and Its Monitoring in the Field A) Attenuation of Local Duck Plague Virus Isolates

Abstract/ Executive Summary:

The local duck plague virus isolates No.3 and 2 were serially propagated in chicken embryos (CE) for 90 and 70 passages respectively by chorio-allantoic membrane (CAM) and allantoic sac (AS) route of inoculation for attenuation and adaptation of the virus isolates in CEo The virus isolates produced 80 to 100 percent mortality of CE with the production of lesions in their body and in CAM from 60th to onward passages of the virus isolates in CEo Such pattern of embryo mortality was found consistent till 90th passege of virus isolates in CEo. The 90th and 70th passaged CE adapted virus isolates No.3 & 2 were inoculated in (i) DE and one week old ducklings and (ii) Ducks of different age groups to evaluate the degree of attenuation and the antigenicity respectively of CE adapted viruses. The CE adapted viruses produced low mortality (00-20%) of DE and no sign of illness in ducklings. These indicate that the virus isolates are attenuated and adapted to CEo. The antisera to attenuated viruses were prepared in different age groups of birds and were subjected to neutralization test to evaluate the antigenicity of the CE adapted and attenuated virus. The antisera to attenuated viruses were found to neutralize the local virulent DP virus isolates and duck plague vaccine (DPV) viruses as indicated by the virus NI that varied from 2.59 to 2.75 log₁₀ DELD ₅₀. This demonstrated that the CE adapted and attenuated viruses are capable of producing antibody in ducks and ducklings.

Objectives:

To attenuate selected virus isolates by adapting in chicken embryos (CE), and 2. To evaluate the antigenic potency of the attenuated viruses.

Methodology:

Virus isolates: Local duck plague virus isolates No.3 and 2 were used in this study. The virus isolates were selected on the basis of the results of the 1st two years studies of this project. Eggs and birds: The ducks, ducklings, duck eggs and chicken eggs were obtained from the local market, BAU Poultry Farm and Poultry flock of the Department of Microbiology and Hygiene. The eggs were incubated at 37°C for the development of embryos and the embryonated eggs were used in this study. Experimental procedures: The local Duck plague virus isolates No.3 and 2 were continuously being propagated serially in CE by CAM and AS route of inoculation using standard technics of embryo inoculation for its adaptation and attenuation. The lesions and mortality pattern of embyos were recorded daily after inoculation. About 90 and 70 serial passages of the isolates No.3 and 2 respectively were carried out in CE.

Achievement/Findings:

The ducklings inoculated with the CE adapted and attenuated viruses did not reveal any sign of illness even after 2 weeks of inoculation and all the ducklings survived.
