Testing and Quality Control of Poultry Vaccines and Its Monitoring in the Field

Abstract/ Executive Summary:

A large number of chickens in a village and BAU poultry farm were vaccinated with Newcastle disease (ND) vaccines (B₁, and K-vaccine) as recommended for field use. The sera of the vaccinated birds at different period of post-vaccination were subjected to Hemagglutination inhibition (HI) test to measure the antibody level in birds against ND. Shelf life and keeping quality of ND vaccines following storage at-20°C and at room temperature (15°C-22°C) were studied by infectivity assay. All the sera of birds tested at 30 weeks after K-vaccination revealed HI titre of 80 and above and such titre persisted in 90% and 60% of the birds till 32 and 34 weeks of post-vaccination respectively. The infectivity assay of the lyophilized B, and K-vaccine demonstrated a minimum EID₅₀ titre of 10^6.7 and 10^5.2respectively per 0.1 ml of vaccine after 30 months of storage at -20°C. A minimum EID₅₀ titre of 10^6.5and 10^5.0 per 0.1 ml of B, and K-vaccine respectively was found after storage at room temperature (15°C-22°C) for a period of 16 days. During the storage period the potency of vaccines was found to maintain. No bird was found affected with ND in the project area following their vaccination with ND vaccine.

Objectives:

1. Studies on the shelf-life (stability) of these vaccines in the laboratory and their keeping quality during transport and exposure to environment, 2. Monitoring the efficacy of the vaccines in experimental and field uses, and 3. Surveillence of diseases in the experimental area of the field.

Methodology:

Shelf-life of vaccines: Determination of the shelf life of the lyophilized B, and K-vaccines after storing in the deep freeze at-20^oC started in the last year of the project work was continued in this year also. The virus titre of each lot of each vaccine was measured every 2 months interval starting from 22 months till 30 months of storage using infectivity assay method. The 15 vials of each of the two lots of lyophilized B, and K-vaccines were left in the laboratory in winter months at room temperature (15°C-22°C) for 18 days. The virus titre of each lot of vaccine was measured before placing at room temperature and at every two days interval during the observation period. Isolation and identification of Fowl pox and pigeon pox virus: Samples such as scabs and scrappings from the pox lesions of the head region of birds were collected from naturally infected chickens and pigeons of rural areas. From the specimens the fowl pox virus and pigeon pox virus were isolated by using standard technics of CAM route of CE inoculation. The CAM of CE revealing pock lesions were collected and stored at -20^oC for future use. The isolated fowl pox and pigeon pox viruses were identified with the help of Neutralization test by using known antisera prepared in chickens against the respective known viruses. The chickens and pigeons were inoculated with the isolated viruses to produce the disease experimentally and for identification of viruses. Surveillance of diseases in experimental village: This was performed by frequent visit to the experimental village and information obtained from the owner of birds about the incidence of ND. As regards other ailments of birds such as parasitic and/or any other suspected infectious disease, appropriate measures for treatment and/or control of the disease was suggested to the respective farmer. If any dead birds could be procured during a visit to village, these were subjected to post-mortem examination for diagnosis of disease.

Achievement/Findings:

During the period under report no epidemic of ND was encountered in the project village. However, incidence of Ascariasis was detected during post mortem examination of birds found dead and collected from village during visit by the project personnels.
