Investigation on Chronic Respiratory Disease in Chickens - Part Iii

Abstract/ Executive Summary:

Serological investigation was conducted by RSP test to detect the prevalence of Mg in birds of different breeds and age groups. It was observed that the birds possessing antibodies against Mg did not shed the organism either through cloacal or respiratory route. Investigation on non- specific serological reaction to Mg was also performed. The results indicated that chickens developing HI antibodies after administration of vaccines prepared with mesogenic strain of NDV showed positive reaction with RSP test using Mg-antigen.

Objectives:

Investigation on isolation of Mg from chickens and also on the role of Newcastle disease viral (NDV) antibodies on RSP test.

Methodology:

Tissue specimens and their source: The following tissue specimens were colleceted for presence of Mycoplasma in chickens: i) Respiratory tissues including trachiea, lungs, air-sacs, turbinates and infra-orbital sinus materials. ii) Colacal swabs from live birds. iii) Respiratory swabs from live birds: These tissues were collected aseptically from BAU Poultry Farm, Central Disease Investigation Laboratory (CDIL) Dhaka. Newcastle disease viccine: Mesogenic Iyophillized vaccine of Kornrnarov (K) strain of Newcastle disease virus (NDV) as prepared by the Poultry Biologics Unit of the Department of Microbiology and Hygiene, BAU Mymensingh was used. Administration of the vaccine: Reconstituted vaccine at the dose rate of 1 ml/bird was administrated intramuscularly in eight birds. Two birds were kept as unvaccinated contro. The birds were maintained separately with adequate food and water during the experimental peirod. Sera samples were collected from the birds prior to vaccination and after 15,21 28,60 and 90 days post vaccination. Test: The sera samples were tested by RSP using Mg-antigen and by hemagglutination inhibition test using homologous viral antigen.
