Pathogenesis of Humpsore (stephanofilariasis) in Cattle Ii. Stephanofilaria Assamensis Its Vectors and Transmission

Abstract/ Executive Summary:

A total of 1,658 flies (1,386 in ethyl acetate and 272 in normal saline) were examined after having caught from humpsore for identification and detection of Stephanofilaria assamensis microfilariae. During the examination, 9 species of flies, viz., Musca conducens, M. sorbens complex (M. vetustissimal, M. ventrosa, M. formosana, one of Family - Ephydridae, Hematobia exiqua, Brontea sp., Stomoxys sp. and Morellia hortorum were identified. Microfilariae of S. assamensis were detected in 3 species which were M. conducens, M. ventrosa and H. exiqua. The highest recovery were noted in M. conducens. In experimental transmission study, out of 3 apparently healthy cattle with artificially made scarified wounds, 2 exhibited development of humpsore within 3 months. These animals were allowed to graze in nature with 2 humpsore affected animals and another 3 apparently healthy animals as control.

Objectives:

1. To examine and identify different species of flies infesting humpsore with a view to determining the vector(s) and also to observe experimental transmission of the diseases.

Methodology:

During the period from July, 1991 to May, 1992, a total of 1,658 flies were examined after having caught directly from the humpsore. All of them were collected from the adjacent villages to and from the BAU campus. Flies were collected in ethyl acetate (killing agent) and normal saline (1 ,386 flies in ethyl acetate and 272 in normal saline). All were examined with magnifying glass and dissecting microscope for identification and classification. A total of 272 flies collected in normal saline were dissected and examined under microscope for detection of microfilariae of Stephanofilaria assamensis. For this, head, thorax and abdomen of different type of flies were examined separately. Besides, 8 cattle, 2 of which affected with humpsore and 6 apparently healthy were selected for experimental transmission study. Sharp and scarified wounds were made artificially in 3 of the 6 apparently healthy cattle on the hump regions and the rest 3 served as control. Sharp wounds were made in 1 st and 2nd and scarified wounds in 3rd and 4th trials. All 8 animals were kept together to facilitate development of humpsore.
