In Vitro Performance of Five Standard Sweet Potato Cultivars (ipomoea Batata L.)

Abstract/ Executive Summary:

Apical meristems of five standard sweet potato cultivars namely, Daulatpuri, Tripti, Kamalasundari, BARI SP-4 and BARI SP-5 were cultured on MS medium supplemented with thiamine HCl 0.1 mg/l, glycine 0.5 mg/I, pyridoxine 0.5 mg/l and nicotinic acid 2.0 mg/l revealed that the highest 70% calli were developed for Tripti. Of them, 31 % were regenerated into plantlets. Maximum size of callus was 17 mm in diameter for Daulatpuri which was equivalent to 700 mg at 90 days. The plantlet showed very slow rate of growth having maximum 40 mm height with four leaves at 90 days for Daulatpuri and minimum 25 mm for BARI SP-4 and BARI SP-5 and two leaves in Kamalasundari.

Objectives:

To study the in vitro growth potential with excised meristem of five standard sweet potato cultivars.

Methodology:

Pre-mature vines from 30 days old plants of five standard sweet potato cultivars such as Daulatpuri, Tripti, Kamalasundari, BARI SP-4 and BARI SP-5 were used to isolate meristem. The plant materials were prepared for meristem isolation following the method described. MS organic salts supplemented with thiamine HCl 0.1 mg/l, glycine 0.5 mg/l, pyridoxine 0.5 mg/l and nicotinic acid 2.0 mg/l and sucrose 3% were used. Meristem of about 0.5 mm length were excised aseptically and inoculated into 25 x 150 mm culture tubes containing 10 ml of media each. As the sweet potato cultures are highly callus-forming, a second experiment was set in search of developing direct shoot and root, using isolated meristem on MS salts and different concentrations of Kn (0.1, 0.5, 1.0, 2.0, 3.0); NAA (0.5, 1.0, 2.0, 4.0, 8.0); BAP (1.0, 2.0, 4.0, 8.0, 6.0); IBA (0.1, 0.5, 2.5, 5.0, 10.0) and IAA (0.2, 0.8, 3.2, 5.0, 7.5). The isolated meristem were cultured in environment as stated previously. The developed calli were tried to culture on MS medium with supplements as of experiment 2. In the control treatment, only MS medium was used. Data on different characters of proliferation are recorded and analyzed using LSD.
