In Vitro Propagation of Strawberry Through Meristem Tip Culture

Abstract/ Executive Summary:

Disease free strawberry (Fragaria vesca) plantlets were regenerated from proliferating meristem tips in MS medium. For shoot regeneration 2.5 μM/l BAP was found most suitable. Only half strength MS without auxin was found effective for root induction. The successfully established plantlets in the field produced fruits.

Objectives:

To develop a protocol for mass propagation of virus free strawberry plantlets through meristem tip culture.

Methodology:

Shoot tip explants were selected from developing runners of Cornel Tropical type cultivar grown under field condition at the Bangladesh Agricultural Research Institute, Joydebpur, Gazipur. Actively growing 1 cm long runners were taken and rinsed with tap water for 30 minutes. The explants were surface-sterilized in pure commercial bleach (30% NaOCl) with two - three drops trix for 30 min and rinsed three - four times in sterile distilled water. Meristem tips (about 5.0 mm) containing three - four leaf primodia were carefully dissected from the shoot tip using sterile scalpel under stereo-microscope in the laminar flow cabinet. After dissection single meristem tips were placed in MS medium supplemented with different concentrations of BAP (0, 1.0, 2.5, 5.0 and 10.0 μM/1). The incubated cultures were kept at 25 ± 1 ºC under 16 h photoperiod with a light intensity of 2000 lux under white fluorescent tubes. The pH of the medium was adjusted to 5.7 before autoclaving. The material was subcultured four-five times at 30 days interval in the same fresh medium. The data on the number and length of shoots were recorded. The elongated and vigorously growing shoots were placed in half strength MS medium supplemented with 0, 1.0, 2.0 and 3.0 μM/1 IBA for root induction. Number of days required for root induction, per cent root induction, number and length of root were also recorded. After rooting the plantlets were transferred in small plastic pots containing a mixture of sterile soil and sand for hardening. Finally, the plantlets were transferred to the field for further growth and fruiting.
