Cross Pollination Techniques in Lentil (lens Culinaris Medik) Under Field Condition

Abstract/ Executive Summary:

The flowers of lentil are small and fragile which makes artificial crossing in lentil difficult resulting In poor success. An experiment was conducted to improve crossing method with the ultimate goal of producing the maximum number of F₁ seeds of lentils at the Pulses Research Center, BARI, Ishurdi, Pabna during 2000-01. Four combinations of crossing and three periods were selected in order to get the maximum pod set. Although non-significant difference was observed among the combinations, emasculation in the morning and pollination in the evening resulted in the high rate of success in pod setting. The periods showed significant differences. The crossing done between 5-14 January resulted in maximum pod set when the average maximum and minimum temperatures were 21.9° and 7.9°C, respectively meaning that environment had profound effect on artificial crossing in lentil.

Objectives:

To improve the crossing method with the ultimate aim of getting the maximum number of F_I seeds under Bangladesh situation.

Methodology:

This experiment was carried out at the Pulse Research Center of BARI, Ishurdi, Pabna during Rabi season of 2000-2001. The four parents used for hybridization were (i) L-5, a local line with white flower and non-pigmented seed, (ii) ILX 113-55 (ICARDA), with white flower and pigmented seed, (iii) BLX 8405-56 (Local cross), characterized by purple flower, pigmented and spotted ash coloured seed, and (iv) ILX 87247 (ICARDA), having purple flower, non-pigmented and brownish seed. Among the four lines L-5 and BLX 8405-56 were indigenous and the rest two were exotic. Emasculation and pollination were done during morning (07:00-11:00 hrs) and evening (14:30- 17:30 hrs), respectively. Crosses were made following four combinations of emasculation and pollination. The four combinations were: (A) simultaneous emasculation and pollination in the morning; (B) simultaneous emasculation and pollination in the evening; (C) emasculation in the morning and pollination in the evening; and (D) emasculation in the evening and pollination in the next morning. The hybridization works were done by two breeders (I and II). In addition, the above combinations of emasculation and pollination were independently attempted by two breeders. The flowers were chosen for emasculation when the tips of the petals reached 50 to 75% of the length of the sepals. During the emasculation process, the flower bud was held between the thumb and forefinger keeping the keel in upper side. Care was taken so that the peduncle did not bend or twist. Sharp-nosed forceps were used to make an incision on the dorsal side of the keel and all the 10 anthers were removed with the forceps. Care was also taken to ensure protection of the stigma from any type of injury. The forceps was dipped in alcohol for every pollination to prevent contamination in the process of crossing. Emasculated buds were tagged with colored threads for identification. The pollen grains were collected by inserting the forceps through the dorsal end of the keel of the fully open flower to the ventral side. Pollen grains adhered to the forceps was transferred to the stigma of the emasculated buds. The hybridized pods were collected and the seeds set were counted at maturity.

Achievement/Findings:

The periods showed significant differences. It is suggested to cross- pollinate lentil in early morning when the temperature remains low for higher success rate.
