Thin Layer Chromatogrphic Study of Methanol Soluble Compounds of Corchorus Olitorius, C. Capsularis and Their F1 and F2 Hybrids

Abstract/ Executive Summary:

Thin layer chromatographic (TLC) stud y of methanol soluble total and nitrogenous compounds were carried out to prove that the Ft and F2 progenies of the cross between Corchorus olitorius and C. capsularis were genuine hybrids. In the present chromatographic study, it was observed that the hybrid progeny differed in both total and nitrogenous compounds from the parents in respect of number. length and intensity of spots. The TLC study revealed that the progenies which resulted from the above cross were genuine hybrids.

Objectives:

To make the above cross with more care and combining the techniques so far advocated by the above workers and carry out TLC test o f the methanol soluble total and nitrogenous compounds.

Methodology:

Seeds of C. olitorius var. 0-4, C. capsularis var. D-154 and their F1 and F2 hybrids were used in the present investigation. The seed s (250 g) of the above materials were powdered with the help of mortar and pestle. The powdered seeds were then taken into a clean conical flask and methanol was added just to cover the powdered seeds. Extraction was carried out over a period of 48 hours with occasional shaking and stirring. The extract was filtered off and kept in the refrigerator for use. The methanol extract was analysed by ascending one dimensional thin layer chromatography using different solvent systems namely, chloroform : methanol, ethyl acetate : methanol and acetone : methanol. A small drop of the extract was applied at 2 cm above the lower edge of the activated silica gel-G plate (20 x 5 cm) with the help of a fine capillary tube. The spotted plate was then placed in the chromatograhic tank containing selected solvent system. When the developing mobile phase reached the pre marked point, the plate was taken out and dried in the air. The properly developed and dried plate was treated with vanillin-sulphuric acid and heated at 11⁰C for 15 minutes to detect the total number of component (s) separated. For detection of nitrogenous compound(s) the properly developed and dried plate was sprayed with modified Dragendroff's reagent and heated at 11⁰C for 15 minutes

Achievement/Findings:

In the present investigation two chromatograms were run in different solvent system with different proportions of the chemicals making up the solvent. In the first, suitable solvent system was acetone and methanol in the proportion of 6: 4 for detection of the total compounds. The purpose of using the first was to detect the over all difference of compounds in the two parental species and their F1 and F2 progeny by com paring their Rf values. The objective of running the second chromatogram was to detect whether the two species and their F1 and F2 hybrid derivatives differed in respect of nitrogenous compounds. In this chromatographic study, the solvent system that proved best was methanol only. In the present chromatographic study, it was observed that the hybrid progeny differed in both total and nitrogenous compounds from the parents in respect of number. length and intensity of spots. The TLC study revealed that the progenies which resulted from the above cross were genuine hybrids.
