Effect of Alternaria Infection on Husk Constituents in Mustard

Abstract/ Executive Summary:

The level of total sugar, total phenol and ascorbic acid in the husk of two alternaria blight tolerant varieties (Binasarisha-1 and Binasarisha-2), two advanced lines (M- 91 and M-287) and a susceptible variety (Sampad) of mustard were determined at different grades of infection. The constituents were higher in tolerant varieties than in the susceptible ones. With the increase of level of infection the constituents were significantly decreased. Proneness of crop varieties to pathogenic infection were determined by the status of these three biochemical constituents.

Objectives:

To investigate the changes in biochemical constituents (total sugar, total phenol and ascorbic acid) of mustard pod due to infection by Alternaria spp. and its relation to disease proneness of mustard.

Methodology:

Two Alternaria tolerant varieties viz., Binasarisha-1 and Binasarisha-2, one susceptible variety named Sampad; and two advanced lines of BAU M-91 and M-287 were selected in this study. Infected husks were grouped in five on the basis of spots number. Group-1 contained up to 6 spots (T₁), group-2 contained 7 to 25 spots (T₂), group-3 contained 26-50 spots (T₃), group-4 contained 51 to 75 spots (T₄), group-5 contained 76 and above (T₅). The samples were collected from Bangladesh Agricultural University Farm, during the winter season of 1991. The experiment was laid out in a split-plot design and replicated thrice, keeping five varieties/lines in the main plots and the five groups (T-1, T-2, T-3, T-4 and T-5) in the sup-plot. One hundred grams of infected samples were dried and ground to powder. Five grams of each powdered sample were plunged in 80% boiling ethyl alcohol (@ 10 ml/g) for 8 minutes and then cooled in running water. The residues of the extract were again boiled in 80% ethyl alcohol (@ 5 ml/g) for three minutes. Both the extracts were filtered and the total volume of the filtrate were finally raised to 50 ml. This extract contained total soluble sugars and total phenol.

Achievement/Findings:

The synthesis of total sugar, total phenol and ascorbic acid depends on the inherent characters of plants and these constituents discourage the disease development in the plant body. The advanced line M-287 is promising to be selected as tolerant to alternaria blight.
