In Vitro Propagation of Haldu (adina Cordifolia Roxb.)

Abstract/ Executive Summary:

Haldu (Adina cordifolia Roxb.) is a naturally grown tree in moist deciduous forests of Bangladesh and has diversified uses. But the population of the species has become threatened due to over exploitation. The large scale propagation of this species is quite difficult through sexual and asexual methods. So to rescue this important species from going to be threatened, attempts were made to develop an efficient protocol through in vitro culture that would maintain and propagate it in large scale. The shoot tips were used as explants and culture established in MS media supplemented with different concentration of BAP and combination of BAP & NAA 0.5 mg/l. It was observed that MS media supplemented with BAP 1.0 + NAA 0.5 mg/l was more effective for shoot production and development. The present study also showed that IBA was more suitable than NAA where IBA 1.0mg/l was found more effective for root induction. After sequential hardening, rooted plantlets were successfully transferred to soil and the plantlets were grown luxuriously.

Objectives:

To develop an efficient protocol through in vitro culture that would maintain and propagate this important species in large scale to rescue it from going to be threatened.

Methodology:

Growing shoot tips as explants (size about 0.8 - 15 mm long) were collected from adult trees. These were washed first under running tap water for 30 minutes and treated with 1% Tween-20 for ten minutes followed by repeated rinsing with autoclaved-distilled water. Further sterilization was done under aseptic conditions in a Laminar Airflow Hood with 0.1% (w/v) mercuric chloride (HgCl₂) solution for 10 minutes. Finally, the explants were washed thoroughly with autoclaved-distilled water for several times. The explants were cultured on MS medium (Murashige T and Skoog F, 1962) supplemented with BAP (6-Benzyl Amino Purine) along with 3% sucrose and 0.7% agar. The pH of the media was adjusted to 5.8 with prior to autoclaving (20 min. at 121ºC). The sterile explants were inoculated in sterilized MS medium supplemented with BAP (1.0 mg/l) for sprouting. Then the cultures were incubated in a growth room at 25 ± 2ºC with a photoperiod of 16 hour. For multiple shoot production, the sprouted shoots were sub-cultured in MS medium supplemented with four concentrations (0.5, 1.0, 1.5 and 2.0 mg/l) of BAP and combinations of BAP & 0.5 mg/l of NAA (?-Naphthalene Acetic Acid, auxin). For root induction, four concentrations (0.5, 1.0, 1.5 and 2.0 mg/l) of IBA (Indole-3 Butyric Acid) and NAA were supplemented individually in half strength MS medium. The rooted plantlets were removed from the agar and dipped in MS basal liquid medium without carbon sources for 24 hours. Then the rooted plantlets were transferred in potting media containing organic material free soil. Sugar and vitamin free MS salt solution was used to moistened soil and plantlets. Data were recorded on average number of shoots multiplied, height and leaf numbers, average number of roots, root length and nature of roots.

MS medium containing the combination of BAP 1.0 + NAA 0.5 mg/l and IBA 1.0 mg/l produce more shoots and roots respectively that can help in large scale production of Adina cordifolia in place of natural regeneration.
