Evaluation of Six Blotter Techniques for Detection of the Major Fungal Pathogens in Jute Seeds

Abstract/ Executive Summary:

Six different blotter techniques were evaluated to select the most suitable technique(s) for detection of the three major seed-borne fungal pathogens- Botryodiplodia theobromae, Colletotrichum cochori and Maccophomina phaseolina in jute (Corchorus capsularis L.). Among all the blotter techniques compared, the earthen dish-newsprint technique appeared to be most suitable for detection of the three selected fungal pathogens in jute seeds. The techniques is quick, economic and reliable. With this technique health of jute seeds can be tested within a period of only five days instead of seven days normally required by the recommended blotter technique, The technique costs only Tk. 4.75 (US $ 0.15) for testing a single sample which is about nine times less than that of the recommended Blotter technique with Pyrex glass petridish and standard filter paper Whatman No.1 for routine seed health testing.

Objectives:

To select the most suitable technique(s) for detection of the three major seed-borne fungal pathogens- Botryodiplodia theobromae, Colletotrichum cochori and Maccophomina phaseolina in jute (Corchorus capsularis L.).

Methodology:

Two seed samples designated as sample No.1 and sample No.2 of 0-154 variety collected from the farmers of Mymensinqh Sadar Upazila. having 36.0% and 29.0% germination, respectively were used for the study. Seed-borne infection of the three major fungal pathogens-Botryodiplodia theobromae, Colletotrichum corchori and Macrophomina phaseolina were 4.0%, 70.0% and 3.5% in sample No.1 and 5.5%, 25.0% and 9.5% in sample No.2. In evaluating the blotter techniques, two types of incubating dishes-glass petridish and earthen dish, each with three types of incubating papers namely -Indian filter paper, Khulna newsprint and whatman No.1 filter paper were used. Thus, a total of six blotter techniques were compared. Glass petridishes used were pyrex glass and 9 cm in diameter. Earthen dishes used for the study were locally made by the village potters. The earthen dishes were of two sizes-15cm and 12cm in diameter. Both the types of dishes ware 2cm high and slightly tappering towards the bottom. Discs of all the three types of incubating papers used were 9 cm in diameter. Two hundred seeds taken randomly from each seed sample were used for each of the techniques. Before plating seeds, three pieces of each type of incubating papers dipped in sterile tap water were placed on each dish. Thus, seeds were plated in eight replicate dishes following the International rules for seed Testing (ISTA, 1976). In case of earthen dish, seeds were planted in the smaller (12 cm) dish. After planting, the earthen dishes were covered by a thin transparent polythene sheet with the help of a rubber band. The small dish with the seeds plated, were then placed on the bigger (15 cm) dish containing water. All the dishes, thus plated for each technique, were then incubated in the incubation room at 20±2^oC under an alternate cycle of 12 hours darkness and 12 hours day light in the Department of plant pathology, for five to seven days. During incubation, water was added to the bottom earthen dishes in order to keep incubating papers in the inner dishes moist throughout the incubation period.

Based on the present evaluation, earthen dish newsprint technique appears to be the best among all the blotter techniques compared. The technique is quick. economic and reliable. It costs only Tk. 4.75 (Us $ 0.15) for testing a single seed sample which is about nine times less than that of the recommended Blotter technique with pyrex glass petridish and standard filter paper for routine seed health testing.
