A Reproducible Protocol for the Rege Eration of Two Indica Rice Cultivars Br5 and Br11

Abstract/ Executive Summary:

Effort has been made to find out standard protocols for the C of plant lets in two indica rice cultivars (BR5 and BR11) via calls from mature embryo to facilitate improvement of rice storage protein by using genetic engineering. Calli were initiated in LS medium with 2 mg/I, 2, 4-0 + 3g/l yeast extract and complete plantlets were obtained by transferring these calli to medium containing MSD4 + 0.5 mg/l + BAP + 0.5 mg/I NAA. Storage protein was analyzed and total glutelin and prolamin contents have been estimated for each cultivar.

Objectives:

To observe the total protein contents in the seed of selected rice cultivars and the distribution of rice storage proteins like glutelins and prolamins

Methodology:

Dehusked seeds of BR5 and BR11 were surface sterilized with 0.1% HgCl₂ and inoculated in LS with different combinations of auxin and cytokinin for callus induction and were incubated at 25 ± 2°C under 14 hrs light with an intensity of 2000 lux under florescent tube. The calli were subcultured in different other media for callus proliferation, maintenance and regeneration of complete plant lets. The regenerated plant lets were transplanted successfully to pots containing ground soil and organic manure in the ratio of 1:1 and finally to the field. For the extraction of rice storage proteins, from the mature seeds the following flow sheet was followed, all the steps were repeated three times.
