Comparative Study of Protein and Esterase Profiles of Wild Salt Tolerant Rice with Local and High Yielding Cultivars

Abstract/ Executive Summary:

Protein profiles of a wild salt tolerant rice Porteresia coarctata, a local salt tolerant cultivar of indica rice, namely 'Binnatoha' and a high-yielding indica BR7 were compared by SDS-gel electrophoresis at the leaf and seed level. No major differences were observed in the total number or distribution of the leaf proteins. The BR7 protein pattern showed two extra proteins, one of 80 Kd and the other of 16.5 Kd. A protein of 55 Kd in the BR7 lane also stained darker than the corresponding proteins of P. coarctaia and 'Binnatoha'. However, proteins extracted from 90- day old seeds of BR7 were found to be extensively degraded compared to P. coarctata and 'Binnatoha'.

Objectives:

To detect homology in protein and esterase profiles amongst a local salt-tolerant rice called 'Binnatoha' , a high yielding cultivar, BR7 (both indica varieties of O. sativa) and P. coarctata.

Methodology:

Seeds of 'Binnatoha' were collected from the Sonagazi, Noakhali sub-station of the Bangladesh Rice Research Institute (BRRI). The salt tolerance level of 'Binnatoha' is comparable to that of Nona Bokra and Pokkali (unpublished results), two of the well known salt tolerant varieties of the Indian Subcontinent. Rhizomes of P. coarctata were collected from the marshy land close to the coastal region in Sonagazi. These rhizomes were then planted in normal soil in pots maintained in a net house, and subsequently seeds were collected from the matured pods. Seeds of 'BR-T were provided by BRRI. Leaves were collected at the age of 90 days for BR- 7 and P. coarctata and at 120 days for 'Binnatoha'. Total proteins were extracted from seeds and leaves by crushing in 10 mM Tris. pH 7.8,0.1 % sodium dodecyl sulphate (SDS) and 5 mM B- mercaptoethanol in a mortar and pestle. Phenyl methyl sulphonyl fluoride (PMSF) (10 µM) was added to the extract to prevent degradation of the proteins. The volume of the total supernatant was noted and estimated for protein content using the Microtannin Turbidimetric method (5). Proteins were precipitated from leaf and seed extracts using 5 volumes (7.5 ml) of acetone and 10% trichloroacetic acid at 0^OC respectively. Seed proteins were washed twice with acetone. Precipitated proteins were dissolved directly in SDS-sample buffer pH 6.8. Gel electrophoresis of the protein samples was carried out according to Laemmli. One hundred and twenty microgram of each protein sample was loaded per slot. SDS-polyacrylamide gels were run at a constant current of 20 mA, according to the method of Laemmli. Gels were read off a graph with log molecular weight of known proteins as ordinate and relative mobility as abscissa. Marker proteins were purchased from Sigma.
