Karyotype Analysis and Cma-banding in Channa Straitus and C. Marulius

Abstract/ Executive Summary:

Two fish species Channa straitus Bloch. and C. marulius Ham. were found to possess 2n = 40 and 2n = 42 chromosomes with the TCL of 172.50 and 111.00 µm, respectively. The range of individual chromosome length was 2.50 - 6.0 µm in C. straitus and 2.0 - 3.5 µm in C. marulius. The centromeric formula 7m + 6sm + 4st + 23t was calculated for C. straitus and it was 14m + 8sm + 20t in C. marulius. Numbers of CMA-positive bands were eight in C. straitus and 14 in C. marulius and these all appeared in the terminal regions of the respective telocentric chromosomes . The individual CMA-banded area of C. straitus was larger and thicker than that of C. marulius. CMA bands were found to appear in a pair of dots in each sister chromatid of five chromosomes (out of 14) and only a single dot appeared in a sister chromatid in rest of the chromosomes . This preferential banding pattern indicates the presence of facultative heterochromatin n chromosomes of these two species.

Objectives:

In the present study, two fish species viz. Channa straitus and C. marulius were selected for characterizing their karyotype by both the conventional karyotype analysis and CMA banding method .

Methodology:

Channa straitus Bloch . and C. marulius Ham. locally known as "Shol" and "Gajal", respectively belong to the family Channidae were collected from different fish markets in Dhaka city. These were then reared in a well-aerated aquarium in the Department of Fisheries, University of Dhaka. After sacrificing, the materials were preserved in the museum of the department as voucher specimens. Chromosome preparations were made by "Flame drying" standard methods using kidney and gill cells in the Cytogenetics Laboratory, Department of Botany, University of Dhaka following Kibria et al. For CMA banding, the method proposed by Alam and Kondo(l) was followed with some minor modifications . Briefly, the air dried slides were kept in Mcllvaine's buffer for 30 min. These were then treated with distamycin A (0.1 mg/ml) for 10 min in a humid chamber followed by mild rinsing in Mcllvaine's buffer supplemented with MgS0₄. The slides were treated with CMA (0.1 mg/ml) and kept in humid chamber for 10 min. After rinsing a while, slides were mounted in 50% glycerol and kept overnight at 4°C. These were examined under a Nikon-fluorescent microscope with BV (blue violet) fillter cassette.
