Effects of Infection Time of Macrophomina Phaseolina on Jute Studied in Pot Experiment

Abstract/ Executive Summary:

Macrophomina phaseolina (Tassi) Goid strain MS-6 was artificially inoculated on the stem 6-week old jute (variety D-154 and 0-9897) plant in three different months (May, July and September). Stem inoculation showed that both varieties were moderately resistant in May and September and highly susceptible in July. Plant height, intenoe length, base diameter was lower in inoculated plant than the control. Number of pods and seeds, germination percentage of seeds and seed-weight decreased in inoculated plant. The effect of M. phaseolina due to stem inoculation were more pronounced in July inoculation followed by May and September on both the varieties. Infection percentage of this fungus on seeds obtained from stem inoculated plant was higher in July compared to that in May and September for both the varieties.

Objectives:

The present work has been undertaken to study the infection time of M. phseolina on the two varieties of jute.

Methodology:

Ten pots of 12" diameter each was prepared with soil for each sowing period. Twenty-five seeds were sown in each pot using five replicates for each variety. Each variety was sown in pot at one month interval as on April, June and August for inoculation on May, July and September, respectively. Preparation of the inoculation. For the purpose of inoculation in jute stem, the mass culture of M. phaseolina (MS-6) was made on PDA medium. Circular block (2 mm diameter) was cut from 48 hour old culture with a sterilized block and the inocula -was prepared. Inoculation procedure: After six weeks of each sowing period only ten healthy plants in each pot were kept for study. From these 10 plants 5 for control were selected. The selected plants were sprayed with a fine mist of sterilized distilled water and made ready for inoculation. For inoculation three spots in the stem of plants were sterilized with rectified spirit and pricked with sterilized needle for identification. One block of inocula Plant height, intenode-length and base diameter of stem (average of 5) were recorded before inoculation and after 7 days of inoculation for both the control and treatment sets. Mature pods and seeds were harvested from treated and control plants separately at 3 different times after inoculation. Average number of pods per plant and number of seeds per pod were counted and average weight of 1000 seeds were recorded with an electronic balance. Germination of harvested seeds were recorded after 7 days of inoculation on blotter methods. The results were statistically analyzed by "t" test following Biological Data Analysis (Fry, 1993).

Achievement/Findings:

In the present investigation, occurrence of M. phaseolina was observed more frequently on both varieties of jute in July compared to those inoculated in May and September. The mean humidity, maximum temperature, minimum temperature recorded during July-August, 1996, ranged between 84-85%, 31.80-32.54° C and 26.20-26.83°C, respectively . Such range of humidity and temperature were optimum for the development of pycnidia and sporulation of M. phaseolina .
