Bacteriological Analysis of Cultured Tilapia and the Rearing Water

Abstract/ Executive Summary:

The effect of water quality on the microflora of cultured tilapia ( Tilapia nilotica ) was investigated. The main concern was the bacterial prevalence in rearing water as well as in fish body. Ten to 13 Gram negative bacteria were isolated from the intestine and mucus of Tilapia nilotica and from the rearing water samples. The total viable bacterial counts in the intestine, mucus and rearing water sample were 4.96 x 106 , 3.90 x 106 and 1.45 x 10' Cfu/g, respectively on nutrient agar while on the MacConkey agar plates the counts were 1.29 x 105, 7.03 x 10' and 1.10 x 10' Cfu/g, respectively. The organisms were isolated according to standard procedure and the most frequently encountered organisms were Aeromonas, Plesiomonas, Escherichia coli, Salmonella, Shigella and Klebsiella. Vibrio was not found either in the fish bodies or in the rearing water. It is evident from the study that cultured fishes harbor viable bacterial count in a much safer level.

Objectives:

To investigate the bacterial prevalence in rearing cultured water as well as in fish body.

Methodology:

The fish samples were collected from a cultured pond in Narayangonj district of Dhaka division. The fishes were caught in March 2005 during the morning time between 7.00 a.m. and 8.30 a.m . and kept in a special fish container with ventilation facilities. During sample collection the fishes were examined carefully and fishes with diseases and any physical injuries were sorted out and discarded. At the same time water samples were collected from three sides of the pond according to the standard procedure as described in Melther Toledo DO meter manual. The pH of rearing water samples were measured by Jenway 3010 pH meter, (German). Temperature of the water was measured by a thermometer . Dissolved oxygen (DO), Biological oxygen demand (BOD) was measured following the standard procedure using Meller Toledo AG analytical. Isolation of bacteria from rearing water and mucus and intestine of the fish were done on nutrient, MacConky, Salmonella Shigella agar (SS) and thiosulphate citrate bile salt sucrose agar (TCBS). The viable bacterial count was measured by spread plate method13) on nutrient, MacConkey, SS and TCBS agar. Moreover MacConkey, SS and TCBS agar were used for the isolation of gram negative bacteria, Salmonella , Shigella and Vibrio, respectively. The bacterial isolates were characterized and identified by their morphological and biochemical characteristics(⁴)
