Cellulolytic Microorganisms of Soil in Albizia Procera Forest Plantation

Abstract/ Executive Summary:

An attempt was made to isolate and study the cellulolytic microorganisms from soil in Albizia procera forest plantation . Among the isolates , six were finally selected and iden tified as two bacterial strains ( Micrococcus luteus, Cellulomonas flavigella), two strains of actinomycetes (Streptomyces sp.) and two fungal strains (Aspergillus ochraceous and Trichoderma album). Production of cellulolytic enzymes, extracellular protein, reducing sugar and biomass yield of these strains have indicated that they were potential in cellulolysis.

Objectives:

1. To investigate the cellulolytic microbial population in a deciduous (Albizia procera ) forest plantation at Chittagong University campus and to characterize them for the production of fermentable sugar from cellulosic wastes.

Methodology:

Soil samples were collected in four seasons (October-December, 1991 ; January-March, 1992; April-June, 1992; July-September, 1992) of a year. The forest plantation was divided into three topographical levels such as hill top, mid slope of the hi ll and hill base. The samples were collected in sterilized polyethylene bags from 2.54 to 15.24 cm depth by an auger. Twenty samples were collected randomly from each topographical level of the hill in separate polyethylene bags. Then the samples of each topographical level were mixed together to form a composite sample. Nutrient Agar (NA), Potato Dextrose Agar (PDA) and Czapek's medium (Atlas, 1993) with 2% carboxymethyl cellulose (CMC) were used for the detection of cellulolytic bacteria, fungi and actinomycetes. Morphological, physiological and biochemical charateristics of the selected strains were tested. Field moisture and pH of the soil samples were determined following the methods described in Soil Chemical Anal ysis by Jackson. For screening out the active cellulase producers, the isolates were incubated in 50ml Winstead's medium in lOO ml flask. The bacterial isolates were incubated for two days while the Streptomyces and fungal strains were incubated for 5 to 10 days. The culture supernatant was analyzed for protein, reducing sugar and enzyme activity . For CMC-ase activity, 0.2 ml of the filtrate was added to 4.5ml of 1% CMC prepared in O.lM citrate buffer (pH 4.6) and incubated at 40°C for two hours. For Fp-ase activity, 0.4ml of the filtrate was added to 0.6ml of O. I M citrate buffer at pH 4.6 along with 50mg Whatman No. 1 filter paper strip (1x6 cm) in a test tube and incubated at 40°C for two hours. The amount of reducing sugar was determined by Nelson's modification of Somogy method, measuring the absorbance at 500 nm. Enzyme activity was expressed by the amount of reducing sugar released/ml supernatant/hr. Protein was determined by the method of Lowry et al. measuring the absorbance at 750 nm and using casein as the standard . Bacterial biomass was determined by optical density method at 600 nm while the actinomycetes and fungal biomass was determined by dry weight method.
