Microbial Spoilage of Dried Loita Fish (harpodon Neheius) and Their Preservation by Radiation

Abstract/ Executive Summary:

Microbiological aspects of the spoilage of dried loita fish (Harpodon neheius) and the effect of irradiation for their preservation upto six months were studied. The total viable bacterial count, total coliform count and total fungal count of the loita fish ranged between 7.8x I06 and I.0x 108 , I.0x 102 to 3.0x 103 and 5.0x l03 to 3.0x l03 cfu/g respectively. Dominant bacterial isolates from the dried loita fish were Bacillus cereous, Pseudomonas sp., M icrococcus varians, Micrococccus luteus, Staphylococcus epidermidis and Bacillus alvei. Aspergillus niger and Aspergillus flavus were the major fungal isolates identified from the loita fish. Total bacterial count was found to decrease from 7.8x106 cfu/g to 3.9x 106 , 2.0x 104 , 9.0x I02 cfu/g and undetectable after the treatment of the samples with irradiation doses of 1 , 3, 5 and 7 kGy respectively. Coliform , fungi and total bacteria were completely eliminated by 1 kGy , 3 kGy and 7 kGy of irradiation doses respectively. Total bacterial count was found to increase in non-irradiated sample but the number decreased in irradiated samples during the storage of 6 months.

Objectives:

1. To evaluate the changes of microorganisms associated with irradiated and non-irradiated dried loita fish during storage of six months at ambient condition.

Methodology:

Dried loita fish was collected from five different markets (Kawran bazar, New Market, Kaptan bazar, Polashi bazar and Moulovi bazar) of Dhaka city. One kg of dried loita fish was collected in a sterile polythene bag from each market. Two hu ndred gms of dried loita fish were then packed in a sterile polythene bag and were irradiated at 1, 3, 5 and 7 kGy irradiation doses using gamma irradiator (⁶⁰Co, 50000 Ci). Irradiated and nonirradiated samples were stored at ambient temperature for 6 months and the samples were then analyzed routinely in every month. Standard spread plate technique was used for microbiological analysis. Nutrient agar, potato dextrose agar (PDA) and McConkey agar media were used to culture total viable bacteria, total fungi and total col iform respectively. Nutrient agar and MacConkey agar media were incubated at 37°C for 24 hours and PDA was incubated at 30°C for 48 to 72 hours. Morphologically dissimilar bacterial colonies were picked up with the help of sterile loop and were subcultured to get pure isolates. Pure isolates were then identified by the methods as described in Bergeyfs Manuar of Systematic Bacteriology.
