MD. MOTASIM AHMMED
Examination Roll No. JJ-06/2003, Registration No. 21983 (1995-'96) Semester: January-June, 2004

The biological activities of an ecofriendly insecticide celaflor was evaluated on the mortality, feeding and growth responses of epilachna beetle, Epilachna dodecastigma (Wied.) in the laboratory of the Department of Entomology, Bangladesh Agricultural University, Mymensingh, during January to April, 2004. Mortality response of the test insect to celaflor was assessed by exposing separately all the larval instars to brinjal leaves treated with 0.06, 0.125, 0.25, 0.50 and 1.0% concentration of celaflor. The results showed that all the larval instars of epilachna beetle were highly susceptible to celatlor and the susceptibility gradually increased with increase of concentration and exposure time and decreased with the increase of larval age. The median lethal concentration (LC) for the first instar larvae were 0.51, 0.I 9 and 0.1 I% at I. 2 and 3 days after treatment (DAT) respectively. The higher LD50 values were found at fourth instar larvae and the values were 1.41, 0.80 and 0.40% at I, 2 and 3 OAT respectively. The median lethal time (LD50 ) of eelaflor increased proportionately with larval age and with decreasing insecticide concentration. For feeding and growth responses, the efficacy of celaflor was evaluated by exposing third instar larvae and adult beetles to treated brinjal leaves as continuous treatment. The results indicated that celaflor significantly reduced the food consumption as compared to control. The mean food consumption by larvae and adult beetles were only 64.00 and 8519.60 sq. mm at 0.06% concentration in comparison to 666.80 and 72559.40 sq. mm in control treatment. Celaflor significantly prolonged larval and pupal period and markedly reduced pupal and adult recovery. Adult beetles developed from treated larvae also exhibit morphological abnormalities. It was concluded that celaflor possesses insecticidal, antifeedant and growth inhibiting properties for E. dodecasligma.

1. To study the mortality response of epilachna beetle to celaflor, 2. To study the effect of celaflor on food consumption of epilachna beetle, 3. To study the effect of celaflor on growth and development of epilachna beetle, and 4. To study the effect of celaflor on morphological abnormalities of epilachnu beetle.

The test insecticide celaflor was collected from Dr. Md. Azizul Haque, Professor, Department of Entomology, BAU, Mymensingh. Brinjal leaves were used as food for the test insect. The foods were collected from the Entomology and Horticulture Field Laboratory, BAU, Mymensingh. For the regular supply, a mass culture of the test insect  was established in the laboratory. In this sense, adult beetles were collected from the brinjal field and were reared in the laboratory. They were sexed and confined in paired in petridishes (9 cm dia.) for mating and egg laying. Fresh and good quality leaves of brinjal were supplied daily to meet the food requirement. After oviposition, the adult beetles were transferred in another petridishes leavina the eggs undisturbed for healthy hatching. After hatching. the new borne larvae were transferred in another petridish (9 cm dia.) and reared up to adult emergence. Twenty larvae were reared in each petridish. The newly emerged adult beetles were again sexed and confined in pairs in petridishes for mating and laying eggs. This process was repeated several time to get sufficient amount of larvae and adult beetles. To investigate the efficacy of celaflor, brinjal leaves were treated with different concentrations of celaflor by dipping method. All of the treated leaves were air dried before offering to the insects. In control treatment, leaves were treated with distilled water only. The leaf petiole was covered with water soaked cotton pad to prevent desiccation. The eflicacy of celaflor on the growth and feeding responses  was determined by exposing third instar larvae to food treated with celaflor at 0.01, 0.02, 0.04 and 0.06% concentrations and were exposed the larvae continuously till pupation to treated food. The newly emerged 3rd instar larvae from laboratory stock culture were placed individually in petridish containing treated food. There were 10 replications for each concentration. The treated food was replaced with fresh treated leaves daily. The data on larval period, pupal period, percent pupation, adult emergence and food consumption were recorded. The food consumption for larvae were measured by using square millimeter graph paper. In addition, morphological abnormalities raised due to action of celaflor were also recorded.
