Postharvest Sprouting Physiology and Dormancy Breaking of Potato Microtuber

Abstract/ Executive Summary:

Microtubers of 100 ±25 mg sizes of four potato cultivars Cardinal, Diamant, Heera and Dheera were subjected to different treatments in presence or absence of gibberellic acid (GA) in order to assess post harvest sprouting physiology and dormancy. Except refrigerator storage (4 ±l°C), most of the treatments on the average, showed 2.5% weight loss after 10 days and 24±2% after 60 days. Under refrigerator storage, however physical damage was >40%. Sprouting of tubers started between 34-44 days due to treatments except refrigerator storage. After 60 days 100% tuber sprouted the treatment with 8 mg/I GA₃+30±3° C+12h DL. In other treatments, sprouting varied from 65-88%. Tubers in refrigerator remained almost un shriveled and in other treatments the tubers shriveled at the rate of 1.75 to 3.75 of 1-5 scale. Among the four cultivars, performance of Cardinal and Diamant for most of the parameters proved to be better. The treatment with 8 mg/I GA₃+ 30 ±3⁰ C+ 12h DL appeared to be effective for breaking dormancy of micro tubers.

Objectives:

To investigate the storage behavior of micro tubers of two Dutch and two local HYVs under varied storage conditions and also following gibberellic acid treatments.

Methodology:

Microtubers of 100 ±25 mg weight of two Dutch potato cultivars Cardinal and Diamant and two local HYVs Heera and Dheera, harvested from in vitro culture after 8 weeks, cultured under dark condition, were used. Tubers were subjected to different storage treatments such as, T_l=4±1° C under refrigerator (4±I°C+R); T₂=18±2°C at dark (18±2°C+D); T₃ =24±2°C under 16h light, 3 Kllight intensity (24±2° C+ 16h 3KI L); T₄ =30±3° C under diffuse light (30±3° C+DL) and 10 minutes soaked at five different concentrations of GA₃ (2, 4, 6, 8, and 10 mg/I) and stored at ambient temperature (30±3°C) under diffuse light as T₅= 2 mg/I GA₃+30±3°C; T₆ =4 mg/I GA₃+30±3°C; T₇ =6 mg/I GA₃+30±3° C ; T₈=8 mg/I GA₃+30±3° C; T₉ = 10 mg/I GA₃+30±3° C. Prior to GA₃treatment, 1600 microtubers of four cultivars (400 of each cultivars), about 2-3 sq. cm. skin of each microtuber was peeled out with no. 10 surgical blade in order to better absorption of GA₃ in the tuber. A two factor (cultivar x storage condition) randomized complete block design with four replications was followed to conduct this experiment. Each replication included 20 microtubers. The initial weight of the individual tuber was measured immediately after harvest. The observations were made at 10 day interval. Data collection included per cent weight loss at different days after storing, loss due to other causes, days to first sprouting, sprouted tubers after 60 days, sprout number per microtuber, length of the longest sprout (mm), wt. of sprouts per tuber and tubers physical quality. Tubers physical quality was determined by following an arbitrary 1-5 scale (1= no shrinkage and 5= high shrinkage). The data were statistically analyzed and mean separation was done using LSD.
