Diversity of Starch Granules from Different Plant Sources in Bangladesh

Abstract/ Executive Summary:

Reserve starch was isolated from ten different plants including wheat as standard. These included starch from root , fruit, seed, and stem and modified stem. Starch granules obtained from different sources showed characteristic shapes in scanning electron micrographs (SEMs). These ranged from long elongated granules for Kach kola to triangular granules observed for Pesta alu. Proteins inside starch granules were analyzed by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SOS PAGE ). Markedly diverse patterns of protein bands were observed among the samples. A band of about 60 kDa, which is probably granule bound starch synthase (GBSS), was observed as a main band for each of the samples and the size of this band varied slightly for different samples. The number of bands and their ranges were largest in case of seed (e.g. Kathaler bichi) . This range was observed from 25 to 100 kDa. The proteins found inside the starch granule represent some of the enzymes involved in starch biosynthesis. Identifying and manipulating the activity of these enzymes may allow us to produce starch of different characteristics and uses.

Objectives:

To identification and manipulation of starch biosynthetic enzymes and their activity to produce starch of different characteristics and uses.

Methodology:

Extraction of starch from different plant sources : Starch was isolated from ten different plant species including wheat as standard following the method described by Badenhuizen. Different plant parts utilized for starch extraction included starch from root, fruit, seed, and stem and modified stem. Scanning electron microscopy : Dry starch grains were lightly dusted onto double-sided sellotape on a copper stub, sputter-coated with 30 nm gold , and observed at 15 kV in a Jeol 6400 scanning electron microscope . Analysis of the granule bound proteins : Starch extra-granular proteins were removed from the starch samples extracted from various sources and the intra-granular proteins, which are supposed to be involved in starch biosynthesis, were analyzed after SDS polyacrylamide gel electrophoresis (SDS PAGE ) as described by Rahman et al.

From the results it may be concluded that each starch granule possesses its characteristics size and shape, which depend on the species of the plant; have similar band patterns for different sources of same category of plants and plant parts; and some of the intra-starch granular proteins may be novel and could give new information about starch synthesis.
