Due to some barriers viz. highly heterozygous and cross pollination nature of genus, paucity of information about inheritance pattern of yield and its contributing traits limits the improvement through conventional breeding methods. Invitro techniques such as tissueculture and organ culture offer plant breeder to improve geneticpattern. Biotechnology provides new scope in sericulture to produce virus free sapling and to establish homogenous clone garden from where clone can be supplied to sericulture farmers through extension, exchange of disease free genetic resources of mulberry and their conservation invitro condition through plant tissue culture techniques also provide new opportunity to the breeders to improve this unique plant of sericulture. During the project period standardization of media, media component and optimization of explants and media sterilization were completed. MS media were found more effective 5.50-50.70 pH, 30%sugar/litre and 8%/litre of agar were found optimum for micro propagation. BM-3 was observed more responsive than BM-1 & BM-2 in root shoot proliferation. MS media supplemented with 1mg (BA)/L and 1mg (NAA)/L were found more effective for shoot and root proliferation respectively.During the project period 6 elite mulberry varieties have been collected which can be utilized in the breeding programme for improvement of this unparallel of sericulture.

Variety and Species
A. Identification of few (5-7) elite mulberry genotypes from the heterogeneous population in Bangladesh;
B. Identified and selected mulberry genotypes must have the intrinsic qualities like, quicker and robust growth habitat, high nutritional quality of leaves for healthy silk warm rearing, disease resistance as well as drought resistance that can survive in prolonged summer;
C. Cloning of these elite genotypes to develop clone garden for breeding and large scale propagation program;
D. Large scale propagation of clones through vitro techniques will be considered;
E. Collection of germplasm from neighbouring countries, their maintenance and conservation through in vitro techniques;
F. Micro-propagation and clonal multiplication of difficult to root elite genotypes.
1st Year:
Experiment-1: collection of germplasm- Initial contact will be made with different countries,with diffenent research institutes working on mulberry propagation, genetics, breeding and biotechnology.
Experiment-2:A) Identification and selection of elite genotypes from farmers field.
B) Close observations of the exotic genotype S1 woukd be made to do further selection from within the genotype. Genotype S1 belongs to Morus sp is the best introduced one which is widely cultivated all over Bangladesh for its high yield and nutritious quality.
Experiment-3: A) Through effort will be made to develop the protocol of Invitro cloning with elite genotypes of mulberry. Standardization of culture condition composition, carbon, nitrogen requirement and other aspects of the culture condition will be made.
B) Nature and type of explants will be studied and a monogram will be prepared for explants nature responding in vitro culture.
C) Effort will be made to develop Somaclones through callus culture. E) Effort will be made to induce invitro rooting of difficult-to-root elite genotypes so that a good number of elite genotypes canbe established in the field.
2nd Year:
Experiment-1: A) If optimistic responses are found from the donor countries than an in vitro gemplasm bank and a clone garden will be established with the elite genotypes through in vitro techniques.
B) Nutritional status of test-tube plants will be furnished with different bio-chemical tests.
Experiment-2: Well developed micropropagation system would be developed t raise plenty of elite genotypes for commercial propagation.
Experiment-3: Invitro conservation of genetic resources of elite genotypes of mulberry for indefinite period through the adjustment of organic compounds growth regulators and growth chamber conditions.
Experiment-4: Maintenance and conservation of clone garden of mulberry for future programmes.
Experiments-5: If somaclones found they will be studied and multiplied through invitro techniques.
Experiment-6: Micro propagation of large number of elite genotypes forfarmers.
