Antibacterial Activity of Methanol Extract Channa Marulius Hamilton (giant Sake Headed Fish)

Abstract/ Executive Summary:

Methanol extracts of the giant snake head Gozar fish, Channa marulius H amilton has been tested against four human pathogenic organisms such as Escherichia coli, Pseudomonas spp., Salmonella spp. and Shigella sonnei. Among the four organisms. the highest anti -microbial activity was obtained against Escherichia coli. On the other hand, no acti vity was found for methanol extracts against Pseudomonas spp . The minimum inhibitory concentration (MIC) values against Escheriachia coli, Pseudomonas spp. , Salmonella spp. and Shigella sonnei were found to be 4. 64, 64 and 32 µg/ ml , respectively.

Objectives:

In this study, aqueous methanol extracts of the Giant snake head fish, Channa marulius Hamilton was screened for their in vitro activity against some terrestrial pathogens (Gram-positive and Gram-negative bacteria).

Methodology:

The fish Channa marulius was collected in the month of February 2004 from Municipal Bazar , Kushtia, Bangladesh. The fish was identified by the Bangladesh Fisheries Research Institute (FRI) Cox 's Bazar, Chittagong. After collection, the fish was cut into small pieces, 80 gm of fish was weighed with the electric balance and transferred into SOC ml conical flask. Then 400 ml of methanol was added into it. It was closed by foil paper anc kept in dark for seven days. The crude methanol extracts were then filtered and the extracts were placed in a rotary vacuum evaporator to concentrate. Antibacterial screening: Four organisms, (three Gram-negative i.e. E. coli, Salmonella spp. and Shigella sonnei and one Gram-positive i.e. Pseudomonas spp.) were used in the present study to determine the antibacterial activity of the crude extracts in vitro antibacterial activities of the test samples were carried out by disc diffusion method (Bauer et al. 1966, Barry 1980). In the disc diffusion method, nutrient agar (High media, India) was used as culture media and the discs were placed aseptically over the bacterial culture on nutrient agar plates and incubated at 37°C for 24 hours. After incubation for 24 hours, the zone of inhibition around the discs was measured by millimeter scale. Discs were impregnated with each treatment and control was assayed on duplicate agar medium plate for E. coli, Salmonella spp., Shigella sonnei and Pseudomonas spp. The antibacterial acti vities were determined by measuring the diameter of the zone in mm. The experiment was replicated two times to confirm the reproducible results. Sterile, blank paper discs impregnated with only sterile methanol served as negative control each time. Standard nalidixic acid (30 µg/disc), ciprofloxacin (5 µg/disc) and erythromycine (15 µg/disc) were used as positive control for comparison of the antibacterial activity. Minimum inhibitory concentration (MIC) value of the extract of the Gozar fish was determined in present study following the serial dilution technique according to Reiner (1982).

Achievement/Findings:

In the present study, the MIC values of methanol extracts of Gozar fish against E. coli, Pseudomonas spp., Salmonella spp. and Shigella sonnei were found in 4, 64, 64 and 32 µg/ml, respectively . More detailed work is needed to improve the extraction procedure of Gozar fish and to determine its antibacterial activity for further therapeutic development and to provide an additional weapon in the overall strategy of disease control.
