Studies on Bacterial Isolates and Antibiogram Profile of Milk from Bovine Clinical Mastitis

Abstract/ Executive Summary:

The study was therefore aimed to identify the prevailing causal bacterial organisms of clinical form of bovine mastitis and carrying out in vitro antibiogram studies for their resistancy or sensitivity to commonly used antibiotics. For this purpose, 15 clinical mastitic milk samples collected from Central Cattle Breeding Station and Dairy Farm (CCBSDF), Savar, Dhaka and discrete areas of Mymensingh. A total of twenty three bacterial isolates were recovered that were Staphylococcus aureus (31%), Escherichia coli (22%), Streptococcus spp. (17%), Bacillus cereus (13%), Pasteurella haemolytica (9%), Arcanobacterium (Corynebacterium) pyogenes (4%) and Salmonella spp. (4%). Among the isolates, Staphylococci were the highest frequency as causal agent. Antibiogram studies for these isolates showed that gentamicin was found to be the most effective drug followed by oxytetracycline, neomycin, streptomycin and the least effective antibiotic were penicillin, amoxicillin and ampicillin. From the prospective, it could be concluded that use of gentamicin, oxytetracycline can give the best results for treating clinical mastitis in dairy cattle of Bangladesh.

Objectives:

1. To explore the causal bacterial agent and to study the sensitivity pattern of different bacteria for formulating appropriate therapeutic measures for clinical bovine mastitis.

Methodology:

Collection of milk A total of 15 clinical mastitic milk samples were collected of which 10 from Central Cattle Breeding Station and Dairy Farm (CCBSDF), Savar, Dhaka and 5 from discrete areas of Mymensingh. The milk was collected in pathogen free sterilized falcon tube in a quantity of 10 ml of each. Identification of organism After primary culture of the organism, all the samples were subcultured in nutrient agar, MacConkey agar, Eosin Methylene Blue (EMB) agar, blood agar, Salmonella-Siegella (SS) agar, Triple Sugar Agar (TSI) agar and in Brilliant Green (BGA) agar. Blood agar media were used for Staphylococcus sp., Bacillus sp., Pasteurellla sp. and Arcanobacterium sp. The primary identifications of the organisms were carried out by observing the different morphological colony of bacteria on different agar plate and staining characterization (Gram’s staining and Leishman’s staining) of organism. Different biochemical test like Carbohydrate fermentation test for dextrose, sucrose, lactose, maltose and mannitol, Methyl red and Vogesproskauer (MR-VP) test, Indole test and Enzyme activity (Catalase ) test were performed for confirmatory diagnosis of the identified organisms. In vitro antibiotic sensitivity test In vitro antibiotic susceptibility testing of the isolated and identified organism were conducted by using commercial antibiotics discs through disc diffusion method. For that, about 700μl of different bacterial suspensions were poured over nutrient agar plates. After even spreading and solidification, different antibiotics discs were carefully placed with the help of a sterile fine pointed forceps. Then the plates were incubated for 24 hours at 37⁰C and the zone of inhibition around each disc was evaluated. The diameter of zone of inhibition was measured in mm and the quantum of sensitivity was interpreted.

Achievement/Findings:
