Ffect of Gamma Irradiation on Indica Rice Callus

Abstract/ Executive Summary:

Regenerated calli of four rice varieties/ genotypes were exposed 1 to 6 Gy of gamma rays to determine their effects on callus growth and plant regeneration capability. Both callus growth and regeneration capacity were found to be decreased with increasing levels of exposure to gamma rays; plant regeneration capacity was found more sensitive to gamma rays than callus growth. The dose 5 Gy of gamma radiation was found to be the inhibition dose (50%) for callus growth and plant regeneration in Binnatoa. Whereas , the inhibition dose for Pokkali and BR 26 was 4 Gy and for BR 16 was 3 Gy.

Objectives:

To investigate the extent of variability on callus production and plant regeneration of indica local rice cultivars at different doses of gamma irradiation.

Methodology:

Rice (Oryza sativa) cv. Binnatoa, Pokkali, BR16 and BR26 were used in study. Matured and healthy seeds of these varieties were dehusked, surface sterilized in 0.1% HgCl₂ for 20 minutes and then sterilized seeds were cultured in MS medium with 1.5 mg/1 2,4-D, 0.25 mg/1 BAP, 30 g/1 sucrose and 7% agar to initiate callus. Calli of 21-day-old were subjected to 7 different doses (0, 1, 2, 3, 4, 5 and 6 Gy) of gamma rays from ⁶⁰Co source. About 30-40 calli were exposed to each radiation level. Irradiated calli were transferred to MS medium supplemented with 0.5 mg/1 NAA and 10 mg/1 Kn. Repeated subculturings were done at 15 days interval for callus proliferation and organogenesis. Three-week old irradiated calli continued to be proliferated and differentiated into shoots. When these shoots grew to about 3-4 cm in height, these were separated aseptically from each other and transferred to freshly prepared rooting medium (half strength MS + 0.5 mg/ 1 IBA) and kept in growth room at 25 ± 2°C. Data were recorded on survival rate, plant regeneration and number of regenerated shoots per callus.

Achievement/Findings:

The results of this study revealed that 4 Gy of gamma-ray might be considered as LD₅₀ for both the rice varieties Pokkali and BR26 . The LD_5o for Binnatoa and BR16 were found to be 5 Gy and 3 Gy, respectively as these varieties showed 50% survivability after irradiation of calli.
