Determination of Health and Quality Status of Farmers’ Stored Lentil Seeds of Dumki Upazila, Patuakhali

Abstract/ Executive Summary:

Sixteen seed samples were collected from the farmers’ houses of Dumki Upazilla under Patuakhali district 15 days before sowing in Rabi 2011. The seed samples were subjected to health and quality tests. The seed borne infection of the samples was detected by Dry inspection and Blotter incubation methods. Purity, moisture content and germination of the seeds were also determined. The moisture content ranged from 10% to 14.20%. The seed samples composed of 84.42-94.23% pure seeds, 60.10-80.51% healthy seeds, 6.33-13.33% shriveled seeds, 3.11-5.67% discolored seeds, 3.21-5.53% partially filled grains and 2.10-5.82% inert matter. The germination percentage ranged from 64 to 84.23%. Six fungal pathogens namely Aspergillus flavus, Aspergillus niger, Penicillium spp., Fusarium Spp., Sclerotium rolfsi and Alternaria were detected from the seed samples. The proportion of healthy seeds was low in most of the samples; consequently the germination percentage was lower than the seed standards fixed by the National Seed Board.

Objectives:

To determine the health and quality status of farmers’ stored lentil seeds of Dumki Upazilla in Patuakhali

Methodology:

Sixteen (16) seed samples (500 g each) were collected from the farmers’ houses of Dumki Upazilla under Patuakhali district at 15 days before sowing in Rabi 2011 where they stored seeds in earthen jar. Moisture content of the seeds of each samples were determined by (low constant oven method where temperature was maintained 103±2ºC immediately after seed collection. Forty (40) g seeds from each sample were taken and purity analysis was done following standard procedure (ISTA, 2003). Seeds were divided into three components viz. pure seeds, other seeds and inert matter. The weight of each component was taken by electric balance and the results were presented in percentage. Germination test was carried out according to ISTA, 2001. From each sample, 400 seeds were placed in water soaked sand in plastic tray. Four replications were used and the tray was kept at room temperature (23ºC). After seven days, normal seedlings, abnormal seedlings and diseased seeds were counted and the results were expressed in percentage.Four hundred seeds were taken randomly from each sample for Dry inspection and categorized into i) healthy seeds ii) shriveled seeds iii) partially filled grains and v) inert matter. Seed heath was determined by Blotter incubation method to detect the seed borne pathogens (ISTA, 2001). Randomly 400 seeds were taken from each sample. The seeds were placed on water soaked three folds blotter paper (Whatman No. 1) in 10 cm plastic petridishes. Twenty five seeds were placed at equal distance in each plate. The seeds were incubated at 20±2ºC under 12/12 hours alternating cycle of Near Ultra Violet (NUV) light and darkness (ISTA, 2001). After seven days of incubation, petridishes were observed under stereo-microscope at 16X and 25X magnifications in order to record the incidence of seed borne fungi.

Achievement/Findings:

The quality of farmers’ stored lentil seeds are not up to the mark of quality as mentioned by National Seed Board (pure seed minimum 95%, other variety maximum 1%, other crop seed maximum 1%, obnoxious weed maximum number 0.08, inert matter maximum 3%, germination capacity minimum 80%, moisture content minimum 10% and diseases 0.0%).
