The higher population was observed in post retting water than that of pre-retting water. Retting speed was accelerated by the addition of microbes and retting period was reduced to almost half.

To study the microbial population and their impact on water during jute retting in certain jute growing areas in district of Pabna and Tangail at pre and post retting period.

Jute retting water was collected aseptically at pre and post-retting condition. When the fibres of the middle portion of the plant became well separated the retting was considered as completed. The water were collected aseptically from different places of Pabna and Tangail. Ten ml of water from each sample was transferred to 250 m1 conical flask containing nutrient broth. After 7 days ot incubation, 10 ml culture from each of these samples were heated in water bath for 20 min at 80°C to limit the study only for spore forming anaerobes. Then the samples were plated out in nutrient agar by using serial dilution method (1 in 105). Representative colonies appearing after 7 days incubation were counted to determine the population of anaerobes. Cultures of anaerobes were incubated in specially made anaerobic jar. For aerobic culture, Nutrient broth of all the samples were directly plated out without prior heating and incubated at 37°C in the presence of air in incubator. Representative colonies appearing after 7 days incubation were counted to determine the aerobic bacterial population. Similarly fungal populations were also determined. For bacterial culture pH of the medium was 7.2 and for fungal culture it was 5.4. Water samples were collected from a depth of 10-15cm below the surface using a 250ml glass stopper bottle with scale. Samples for dissolved oxygen (DO) determination were preserved as directed under oxygen. Samples were preserved in chilled (3- 4°C) condition immediately for biological oxygen demand (BOD) determination. Samples for chemical oxygen demand (COD) were preserved by adding sufficient H2S04 to obtain a final pH of 2-3.

BOD were determined according to and DO were analyzed by Winkler or iodometric method which is a titrimetric procedure based on the oxidizing property of DO and estimated DO in LG range. On the other hand, the 1 dichromate reflux method following APHA was used for the COD determination. DO content of water was measured by DO meter. CO2 and H2C03 alkalinities and free CO2 were determined by titrimetric method. Aqueous methyl orange indicator solution was added to 100 ml water sample and was titrated with standard dilute hydrochloric acid until the end point was found. A blank test was carried out using the same procedure. The blank correction was subtracted from the volume of standard acid used. The corresponding weight of calcium carbonate was then converted into parts per million. To determine the retting efficiency of the culture pieces of 12 cm long jute stem, weighing 40 g were used as retting materials. Some weighing materials were used to submerge the pieces of jute stem in the distilled water of surface sterilized earthen plates. The stem pieces were placed into Sankee containing 800 ml of distilled water. 10 ml of Nutrient broth culture of all samples were then added to each of the Sankee separately. Sankees were covered with another surface sterilized Sankee and kept at room temperature of 29-30°C. Progress of retting was tested aseptically 7 days after incubation. Fibre quality was assessed by "Touch and Feel" method (recommended by Bangladesh Jute Research Institute). In control, no bacterial inoculum was added, only 10 ml of distilled water was added.

The present study focused on the jute retting efficiency of retting water. The retting water containing higher amount of microbes, showed better performance in jute retting than that of retting water with lowest amount of microbes. Information had been generated in this study that the chemical constituents of retting water create the environment favorable or unfavorable for the microorganisms based on the amount of constituents present in retting water. In favorable environment microbial load would be higher and the retting would be better than lower microbial load in unfavorable environment.
