Interspecific Hybridization Between Brassica Juncea and B. Carinata and Study of F1 Hybrid

Abstract/ Executive Summary:

Hybridization was made to see the crossability and to develop hybrids between two Bmssica species, B . j uncea and B . carinata . Cross between Barisari sha - 11 or B. j uncea (2n= 36, AABB) and Carinata-1 of' B . carinata (2 n=34 , BBCC) was only successful when the latter was used as female parents and rate of success was 8.0'%. Morphological characters such as leaf shape, plant height, number of primary branches per plant and beak length of the hybrids (ABBC) were found to be intermediate between the parents. The hybrids had only 19% - 27% pollen fertility but produced higher number of siliquae per plant than the parents. Hybrids were found to have shorter siliqua having lower number of seeds than the parents . Duration of flowering and maturity of the hybrids was found intermediate between the parents.

Objectives:

To see the crossability and to develop hybrids between two Brassica species, B . j uncea and B . carinata.

Methodology:

Barisarisha-11 of B. juncea and Carinata-1 of B. carinata were selected for the interspecific hybridization programme. The experiments were conducted during 2003-04 and 2004-05 growing season at Bangladesh Institute of Nuclear Agriculture (BINA), Mymensingh, Bangladesh. Crossing including reciprocal were attempted between the two selected genotypes (Barisarisha-11 and Carinata-1) under field condition at BINA. Mymensingh, Bangladesh during the winter season of 2003-04. Young flower buds of appropriate size of the female parent , supposed to open in the next morning , were emasculated before anthesis and the remaining opened flowers and immature buds were removed. The emasculated buds were then pollinated ith fresh pollens collected from the male parent and were bagged immediately with thin brown paper bags. A total of 115 crosses were made using Barisarisha-11 as female and Carinata-1 as male parent, but I00 crosses were done in their reciprocal crosses i. e., u sing Carinata-1 as female and Barisarisha-11 as male The siliquae bearing hybrid seeds were collected after proper maturation and were then stored to grow F 1 hybrids in next cropping season. Hybrid plants with their parents were grown at the same location during the winter season of 2004 -05. Pollen fertility tests were performed by microscopic examination of pollen grains stained with 1.0% acetocarmine . Pollen grains were scored as viable if round shape and stained red or non-viable if shrunken and unstained. The ratio of stained pollen to the total was expressed as percentage of pollen fertility. Data on plant height, number of primary branches per plant. pollen fertility, number of siliquae per plant , silica and beak length , number of seeds per siliqua, days to flowering and days to maturity were taken from the hybrids and their parental sample plants.
