Spectrum of Variability in Seed Size and Content of Protein and Boaa in Grasspea (lathyrus Sativus L. ) Germplasm

Abstract/ Executive Summary:

Seeds of grasspea contain an unusual amino acid, B-(N)-Oxalyl-amino-L-alanine (BOAA) Which is responsible for an incurable paralysis of the lower limbs of men, called lathyrism. This study included sixteen local and seven exotic cultivars of grasspea to ascertain the variability and the relationship exis~ing among seed size and content of protein and BOAA in the seeds. Weight of 1000 seeds varied from 4C-67 gm and the exotic germ plasm had comparatively bigger seeds than the local cultivars. The protein and BOAA percent ranged from 24.54 to 34.05 and 0.22 to 2.08, respectively. A significant positive correlation Was obtained only between seed size and protein content ( 0.775) but negative association was obtained between seed size and BOAA. There Was no association between the content of protein and BOAA. Therefore, it may be feasible to develop low neurotoxin varieties by selecting for bold seed size following hybridization of bold seeded and low neurotoxin types with those having small seeds and high neurotoxin.

Objectives:

To determine the variation in seed size and in the amounts of BOAA and protein in the seeds, and also to find out the relationships, it any, among those characters.

Methodology:

Seed samples of about 10 gms of 16 local and 7 exotic cultivars were dried under a constant temperature of 60⁰C for 24 hours. These were ground by "Retsch" grinding machine using 12 mm sieve after removal of seed coat. Paper chromatographic method Was used for quantitative determination of BOAA following the techniques described by Nagarajan and Mohan (967 ). Powdered sample of 2.5 gm Was extracted with 100 rnl of 70% ethanol which was then concentrated to dryness on a steam bath. These were again extracted with 10% isopropanol from which spots were made on the chromatographic paper along with standard BOAA solutions of different concentrations. The paper was developed overnight in a chromatographic chamber using phenol- water (4: 1 ) as de.veloping solvent front The air-dried paper Was then sprayed with 2% ninhydrrn solution for the development of pink colour. For quantitative estimation, the spots of unknown and standard BOAA samples Were cut and eluted in 75% ethanol solution. The concentration of BOAA of unknown samples were calculated from the standard curve by knowing the optical density at 540 nm with a spectrophotometer ( Spectronic 20 ). The convantionot rnicrokjeldahl .rnethod Was adopted for the determination of crude protein. Powdered samples weighing 200 mg were taken in the rnicro-kjeldahl flask along with a Kjel-tab ( 5 gm K₂S0₄ and 0.25 gm -HgO). Then 3 ml of cone. H₂S0₄ and 2 ml of H₂0₂were also added to help digestion which was made in a "Tecator" digestion set at a temperature of 370^oC. These samples were then disti lied with the help of a "Buchi" distilfation unit which was finally titrated with standard HCL to determine the concentration of nitrogen. The crude protein WaS calculated by multiplying the nitrogen content with the factor of 6.25. Weight of 1000 seeds in gm for each of the local and exotic cultivars
