Pachytene Karyotype in Cajanus Cajan and Atylosia Scarabaeoides

Abstract/ Executive Summary:

The individual pachytene chromosomes of the entire complement of C. cajan and A. scarabaeoides were identified on the basis of relative length, arm ratio, chromosome pattern and nucleolar association. The pachytene chromosomes of both the genera were found to belong to the differentiated type. C. cajan possessed four median, five submedian and two subtermianl chromosomes while A. scarabaeoides was characterized by the presence of six median and five submedian chromosomes. On the basis of various criteria employed for the identification of pachytene chromosomes, C. cajan was found to bean advanced type and A. scarabaeoides a primitive relative of C. cajan.

Objectives:

To bring out similarities and differences amongst the pachytene chromosomes of their entire complement on the basis of relative length, arm ratio and nucleolar association.

Methodology:

For pachytene analysis, flower buds of Cajanus cajan and Atylosia scarabeoides were fixed in aceto-alcohol (1 : 3) saturated with ferric acetae. The buds were kept overnight in the fixative and preserved in 70% ethanol. Anthers were stained with 2% acetocarmine. Using a piece of thread, chromosome lengths were measured from the camera Iucida drawings. Chromosome lengths were measured in mm and then converted into microns (µm). Those fractions of mm which were less than 0.5 mm were not considered (except in the case of heterochromatin where it was recorded as 1.0( µm) and those fractions with 0.5 mm or more were read as 1.0 mm for the sake of convenience. The length of individual chromosomes in the table represent the average length of a minimum of five cells. On the basis of length the entire pachytene complements of C. cajan and A. scarabaeoides were divided into three groups: long (50.0 µm and above), medium (49.0 to 30.0 µm) and short (below 30.0µm). For analysis, chromosomes were classified according to the position of the centromere and their arm (short I long arm) ratios. The chromosomes with the arm length ratio less than 0.50 were placed in subterminal (st) the arm length ratio between 0.51 and 0.75 into the submedian (sm), and those with the arm length ratio above 0.75 into the medium (m) group. The chromosomes were numbered in order of their decreasing length.
