An experiment was conducted in the farmer’s house at Sutiakhali village of Mymensingh district to find out the effect of storage containers on the seed health status of BRRI dhan 47 collected from Borobazar, Khulna. Five containers viz. Tin box (T1), Polythene bag (T2), Earthen pot (Motka) (T3), Gunny bag (T4) and Plastic jar (T5) were used. After six months of storage of rice seeds in those containers, Seed health tests were done two times, once at three months of storage and finally after six months of storage period. Moisture, seed germination, normal seedlings and prevalence of seed borne pathogens were recorded as 12.8, 90.67, 86.67 and 27.72% respectively. Associated seed borne pathogens were recorded as Fusarium oxysporum (4.00%), Aspergillus niger (7.25%), Penicillium sp. (8.00%), Rhizopus sp. (8.50%) and Xanthomonas oryzae pv oryzae (0.02%). Percent germination and normal seedlings were found decreased as the storage time is increased. Among all the containers, Tin box had showed the least seed borne infections while gunny bag was found worsen as because Tin box is more impermeable than gunny bag that prevent humidity inside the box. Tin box may be recommend for the farmers to store their BRRI dhan 47 seeds as it is found suitable to maintain good seed health status compared to other tested containers.

  1. To detect the prevalence of seed borne fungi in seeds stored at different containers and
  2. To explore the knowledge of the suitable containers for preserving saline tolerant rice var. BRRI dhan 47.

Two kilogram seeds of BRRI- dhan 47 were collected from Boro Bazar , Khulna. The seeds were tested for two times at three months of interval rather initial health tests. Five storage containers viz. Tin box, Plastic jar, Earthen pot (Motka), Gunny bag and Polythene bag were used in this experiment. The selected containers were filled with seeds and closed with their lids. The polythene and gunny bags were tightened with thread properly and stored at 25±2° C in the farmer's house (Mud wall and tin shed) at Paglabazar near BAU, Mymensingh. Initial moisture content of the seeds were 12.8%. About 200-300g seeds were used per container as required to fill up the containers based on the size. The lids of the containers were placed firmly to make it air tight. The containers were kept on the floor of the house. The health status of the seed sample was tested before storing by Blotter incubation test. Three layers of blotting paper (Whatman No.1) were soaked in sterilized water and placed at the bottom of a 9 cm diameter plastic petridish. Twenty five seeds were placed on the moistened blotting paper in each plate. 400 seeds were tested after three and six months of duration. Petridish containing seeds were incubated at 25±2°C under 12 hours alternating cycles of Near Ultra Violet (NUV) light and darkness for 7 days. After incubation, the seeds were examined under stereo binocular microscope in order to record the incidence of different seed borne fungi. Temporary slides were prepared from the fungal colony and observed under compound microscope if necessary. Appropriate keys were consulted for identification of the fungi. The results were presented as percent incidence for individual pathogen. Germination was counted after 7 days of incubation on each Petridish and observations were made for total and normal seedlings. Moisture determination of BRRI dhan 47 were done at both three and six months of storing of seeds using by Digital Moisture Meter. To obtain pure culture of seed borne fungi hyphal tips were transferred aseptically onto PDA plate by using the flame sterilized tip of an inoculation needle. For Xanthomonas oryzae, bacterial inocula was taken out from infected rice seeds and streaking at NA medium. The plate was incubated at room temperature. For morphological characters colony color and surface textures were carefully studied as described and recorded after 24 hours of incubation in NA medium. Nutrient agar (NA) was prepared as described. Potato Dextrose Agar (PDA) was prepared as described and poured in 500ml glass bottles and sterilized in an autoclave at 1210C, 15 PSI for 15 min. The media were acidified with 30 drops of 50% lactic acid per 250ml medium to avoid the contamination of bacteria. The experiment was laid out in Complete Randomized Design (CRD). A standard computer package statistical procedure (MSTAT) was followed to find out the mean difference among the treatments.

Incidence of most of the field fungi was found to reduce with the increase in storage period. In our study, Xanthomonas was not found when storage duration was increased. May be the strain of Xanthomonas had lost its viability for long time storage of seeds. However, there was no reduction on the occurrence of the storage fungi. Regarding farmer’s conditions and health status of BRRI dhan47 rice seeds, highest germination and percent normal seedlings were observed in Tin box (T1) while Gunny bag showed least effective performance. From the result of this present study it may be concluded that among all the containers, Tin box may be the best container for prevention of seed borne infection and successful BRRI dhan 47 rice seed production that will ensure our farmers of saline area for their food safety and security.
