Five genotypes of cauliflower (Brassica oleracea var. botrytis) were used to screen their in vitro regeneration potentiality and to establish an efficient protocol for each. Different concentrations and combinations of growth regulators were used in MS medium to observe the callus induction ability from hypocotyls explants. Callus proliferation, shoot regeneration and root initiation were monitored respectively. The highest average per centage of callus induction (84%) was observed in MS medium containing 2.0 mgL-1 BAP, 0.2 mgL-1 NAA, 2.0 mgL-1kinetin and 2.0 mgL-1 AgNO3. Among the genotypes used, Snow Queen induced the highest per centage (100%) of callus followed by Early Tropical (96%). For the maintenance of calli, MS + 3.0 mgL-1 BAP + 0.2 mgL-1 NAA + 2.0 mgL-1 kinetin + 2.0 mgL-1 AgNO3 showed the best result. The average per centage of shoot regeneration was highest (56%) in MS + 4 mgL-1 BAP + 0.2mgL-1 NAA + 2 mgL-1kinetin +2 mgL-1AgNO3 among the growth regulator treatments. Among the five genotypes tested, a large variation in shoot regeneration frequency was observed. In case of shoot regeneration, Snow Queen and Early Tropical showed better results (80% and 76%, respectively) than that of other varieties. The average per centage of root induction was highest (66%) in half MS + 0.5 mgL-1 NAA. When the plantlets with sufficient roots were transferred successfully in plastic pots, Snow Queen and Early Tropical survived easily.

1.To study the in vitro regeneration potentiality of five cauliflower genotypes and

2.To establish a reproducible, efficient and rapid regeneration protocol for cauliflower.

Five genotypes of cauliflower (Brassica oleracea var. botrytis) namely, Snow Queen, Early tropical, Shiratama, Maya F1 and Tangail Special Pauslali were used to study different parameters associated with plant regeneration. The seed materials were collected from the local market of Mymensingh town. Among the five genotypes, three were Japanese (Snow Queen, Early Tropical, Shiratama) and two were marketed by two Bangladeshi seed companies (Maya F1 and Tropical). The experiment was conducted in the Tissue Culture and Genetic Engineering Laboratory of the Department of Genetics and Plant Breeding, Bangladesh Agricultural University, Mymensingh, during November 2005 to April 2006. Murashige and Skoog (MS) and half strength MS media were used as culture media. Different concentrations and combinations of plant growth regulators were used with these media as different treatments. Separate stock solutions for macronutrients, micronutrients, irons, vitamins, growth regulators, etc. were prepared and stored appropriately for use.Surface disinfection’s was done using 0.1% HgCl2 with one drop of Tween-20 for ten minutes. Hypocotyl from each germinated seedling was cut into 5-7 mm pieces using sterilized surgical blades and used as explants. MS medium containing different concentrations and combinations of plant growth regulators were used for subculture of both callus and regenerated shoots. The plantlets developed from calli were transplanted to pots containing garden soil, sand and cowdung in the ratio of 1:2:1.To investigate the effects of different treatments and responses of different varieties for callus induction and plantlet regeneration, the collected data were days of callus initiation, per cent callus induction, size of callus, weight of callus, per cent plantlet regeneration, days of shoot initiation, per cent shoot regeneration, average number of shoot per callus, total number of shoots per vial, per cent plant establishment etc. All the necessary data (total, average, per centage, etc.) and measurements were made using different equations. The experiment was set in a Completely Randomized Design (CRD) for scientific interpretation and analysis of data.

The responses of shoot regeneration of five genotypes were different. Among the genotypes, the regeneration performances were found better in Snow Queen (80%) and early tropical (76%) followed by Shiratama (56%). The MS medium supplemented with 4 mgL-1 BAP + 0.2 mgL-1 NAA + 2 mgL-1 kinetin + 2 mgL-1 AgNO3 showed the highest average percentage (56%) of shoot regeneration. Growth regulators x genotype interaction showed significant variation for all the characters. ½ Ms medium without hormone, ½ MS medium + 0.5 mgL-1 NAA and ½ MS medium + 0.5 mgL-1 IBA were used to observe rooting response of regenerated shoots. The highest percentage (80%) of root induction was observed in Snow Queen and the lowest (40%) in Maya F1. Among the three treatments, MS medium + 0.5 mgL-1 NAA was found to be the best (average 66.0 %) for root initiation followed by half strength MS medium + 0.5 mgL-1 IBA.
