Lateral buds from field grown mature female papaya plants were used as explants. Seasonal endophytic contamination was suppressed by shaking propagules for 2 h in 300 mgl-1 rifampicin before surface sterilization. The maximum survival rate was found when MS medium was supplement with both 0.50 mgl-1 BAP plus 0.20 mgl-1 NAA and 1.0 mgl-1 BAP plus 0.20 mgl-1 NAA. The maximum number of explants produced shoots both in MS medium containing 0.50 mgl-1 BAP plus 0.20 mgl-1 NAA and 1.0 mgl-1 BAP plus 0.10 mgl-1 NAA. The average multiplication rate was about 10-fold per culture in MS medium containing 0.50 mgl-1 BAP plus 0.20 mgl-1 NAA. The highest multiplication rate was found from 4th subculture and a decreasing tendency in 5th subculture. The highest shoot length was found from 2nd subculture, which decreased thereafter. Rooting was obtained at a high rate on half strength MS medium supplemented with 1.0 mgl-1 indole-3-butyric acid. Rooted plantlets were transplanted in to soil successfully.

To determine in vitro regeneration of papaya (Carica papaya L.) from mature female plants

The experiment was conducted in the Tissue Culture Laboratory of Biotechnology Division, Bangladesh Agricultural Research Institute (BARI), Joydebpur, Gazipur during June 2004 to March 2005. Lateral buds consisting of the top young unexpanded leaves from field grown female plants were used as explants. The buds were shaken in rifampicin (RIF) at 300 mgl-1 to suppress the bacterial contamination. The buds were then cleaned with soap water and treated for 7 min with mercuric chloride (0.1 %) containing a few drops of Tween- 20. This was followed by 3-4 washings with sterile distilled water. The explants were agitated in sterile distilled water for 2-3 h in horizontal shaker at 160 rpm before inoculation to minimize the flow of latex into medium. The MS basal medium was used with different concentration of BAP (0.1, 0.2, 0.3, 0.4, 0.5, 1.0, 2.0, 3.0, 4.0 mgl-1) and NAA (0.10 - 0.30 mgl-1) for establishment of explants and BAP (0.5, 1.0 and 2.0 mgl-1) with NAA (0.10 - 0.30 mgl-1) for shoot multiplication. Individual elongated shoots were excised carefully and transferred to half strength of MS media with various concentration of indole-3-butyric (IBA) at different concentration (0.10 - 1.5 mgl-1). The PH of the media was adjusted at 5.8 prior to autoclaving and adding 8 g/I agar to solidify the media. The cultures were kept under approximately 2000 lux fluorescent light at 25 ± 1°C with 16 h photo period.
