Effect of Temperature and Ph on the Growth and Sporulation of F. Moniliforme

Abstract/ Executive Summary:

The growth and sporulation of the pathogen F. moniliforme was studied under different temperature and pH level. The objectives of the work was to know the suitable temperature and pH for growth and development of F. moniliforme. The optimum temperature for growth of F. moniliofrme is 25⁰C. But the maximum sporulation was recorded at 30^oC. In case of pH, the widest colony growth and maximum sporulation was recorded at pH 6.

Objectives:

To determine the suitable temperature and pH for the growth and sporulation of F. moniliforme

Methodology:

Temperature: A mycelial disc from an 7-day old culture of the fungus grown in PDA were cut form the edge of the colony. Each disc was transferred to the center of a 20 ml plated medium of PDA in glass petridish. Plates were incubated at 5, 15, 20, 25, 30 and 35^oC in incubator. Colony diameters (mm) were measured on the 3rd, 6th and 9th day of incubation. Sporulation was observed on 9th day. The experiment was laid out in CRD with four replications. Data were analyzed by MSTAT-C. Differences among the treatment means were compared following DMRT. To determine sporulation, 10 ml sterile distill water was added to each culture. The surface growth was scraped gently without scarifying the agar. The suspension was placed in a test tube and shaken properly. Mycelial growth was removed. Dilutions were made to facilitate counting of spore. Number of spores/ml was counted with the help of a hemacytometer.

pH: PDA medium was prepared. Nine 250 ml conical flask were taken and poured about 100 ml melted PDA into nine separate flask. The pH of these nine conical flask were adjusted to seven different pH level such as 4, 5, 6, 7, 8, 9 and 10 with aqueous solution of 1.0% lactic acid and 10% sodium hydrooxide solution. All conical flasks with media were sterilized. For each pH level 20ml PDA medium was poured into each sterilized plate and allowed to solidifying. Five mm diameter of young culture blocks of F. moniliforme placed centrally on the patri plates. All the plates were incubated at room temperature. Colony diameters (mm) were measured on the 3rd, 6th and 9th day of incubation. Sporulation was observed on 9th day. The experiment was laid out in CDR with four replications for each period of measurement and pH level.
