Effect of Nacl on Callus Induction and Subsequent Plant Regeneration in Some Fine Grain and Aromatic Varieties

Abstract/ Executive Summary:

The effect of NaCl salt on callus induction and subsequent plant regeneration in 27 Indica rice varieties(fine grain and aromatic) were investigated. Among the different concentrations of NaCl tested, 2.5 g/l was found better callus formation and plant regeneration. Both callus induction and regeneration response were decreased with the increasing of salt concentration in callus induction medium. In callus induction, significant variation was observed among the salt concentrations used as well as the varieties tested. Plant regeneration response was also significantly varied among the calli obtained from medium containing different salt concentrations. Regenerated plants were successfully established in the soil and produced fertile seeds.

Objectives:

Increase the regeneration capacity of indica type fine grain and aromatic rice varieties.

Methodology:

Twenty seven Indica rice varieties with Japonica type Taipei309 (control) were used in this experiment. All were local rice varieties except, BRRI dhan34, BRRI dhan38 and Philippine Katari. Mature seeds were the source of in vitro experimental materials. Dehusked seeds were sterilized by 70% ethanol foe 3 minutes with gentle shaking followed by 0.1% mercuric chloride for 20 minutes. Sterilized seeds were washed for 5-6 times with sterile distilled water and dried with sterilized filter paper. Media for callus induction based on the formulation of MS basal salt supplement with 0.8% agar, 20 g/l sucrose, and 2 mg/l 2,4-D. Three different concentrations of NaCl salt viz. 2.5 (MS2), 5.0 (MS3) and 7.5 (MS4) g/l were used in the medium to evaluate their effect on callus induction. Six sterilized seeds were plated on each Patri dish and 240 seeds were used for each variety for each treatment. For plant regeneration, MS media supplemented with 0.8% agar, 40 g/l sucrose, 1 mg/l NAA and 1 mg/l kinetin was used. Four weeks old calli were transferred into test tubes (25 cm x 200 cm) containing 8-10 ml regeneration medium. Data on plant regeneration was taken six weeks after transfer of calli in the regeneration medium. Regenerated plantlets were then transferred into small pots for hardening, and then transferred to the earthern pots and kept until maturity. A complete randomize design (CRD) with six replications per treatment for each variety was used.
