Physiological Aspect of Sclerotinia Sclerotiarum: a New Pathogen of Mustard

Abstract/ Executive Summary:

An experiment was undertaken in the Plant Pathology laboratory of Oilseed Research BARI, Gazipur to study the effect of different culture media, temperature, pH of culture media on growth and sporulation of S. sclerotiarum causing white mould disease of mustard. The results presented in table 1 and fig. 1 reveal that the highest radial growth was occurred in plates incubated at 25º C followed by 20º C. At 25º C the per day growth rate was 33.4 mm/day and at 20º C it was 29.4 mm/ day. The results presented in table 1 and fig. 1 revealed that the highest and similar radial growth was occurred in plates having pH 4.0 and 5.0 followed by pH 6.0 and 7.0. From the results presented in table 1 indicated that the highest redial growth occurred in PDA media followed by nutrient agar media and corn meal agar media, respectively. The fungus did not grow on V8 juice agar media.

Objectives:

To study the effect of different culture media, temperature, pH of culture media on growth and sporulation of S. sclerotiorum.

Methodology:

Effect of different culture media on mycelial growth: Four culture media namely, potato dextrose agar (PDA), corn meal agar (CMA), nutrient agar (NA) and V8 juice agar (V8) were used in the study. The mycelial block of the pathogen, cut from the edge of the pure culture grown in potato dextrose agar (PDA), were plated at the centre of 90 mm diameter petri-plates of different media and incubated at 28±1ºC. The diameter of the colonies in petri-plates was measured after 72 hours of incubation.

Effect of different temperature on mycelial growth: To determine the effect of temperature on mycelial growth of Sclerotinia sclerotiarum, the mycelial block of the pathogen, cut from the edge of the pure culture grown in potato dextrose agar (PDA), were plated at the centre of 90 mm diameter petri-plates and incubated at 10ºC, 15ºC, 20ºC, 25ºC, 30ºC and 35ºC. The diameter of the colonies in petri-plates was measured after 64 hours of incubation.

Effect of different level of pH on mycelial growth: Five level of pH viz. 4.0, 5.0, 6.0, 7.0 and 8.0 were prepared in potato dextrose agar (PDA) media by adding 0.1 N hydrochloric acid and 0.1 N sodium hydroxide. The mycelial block of the pathogen, cut from the edge of the pure culture grown in potato dextrose agar, were plated at the centre of 90 mm diameter petri-plates and incubated at 28±1ºC. The diameter of the colonies in petri-plates was measured after 64 hours of incubation.

Effect of different culture media on mycelial growth: From the results it is indicated that the highest redial growth occurred in PDA media followed by nutrient agar media and corn meal agar media, respectively. The fungus did not grow on V8 juice agar media.

Effect of different temperature on mycelial growth: For temperature effect study, the highest radial growth was occurred in plates incubated at 25º C followed by 20º C. At 25º C the per day growth rate was 33.4 mm and at 20º C it was 29.4 mm. Slight growth was observed at 15º C and 10º C but at 30º C and above temperature the pathogen did not grow.

Effect of different level of pH on mycelial growth: The highest and similar radial growth was occurred in plates having pH 4.0 and 5.0 followed by pH 6.0 and 7.0. At pH 8.0 the vegetative growth was the lowest.
