Assessment of Rice Cultivars/lines for Low Amylose -2004

Abstract/ Executive Summary:

Rice is classified into glutinous and non-glutinous depending on amylose content. Glutinous rice contains no or very little amylose and non-glutinous rice contains more than 10% amylose. Non-glutinous rice which contains >20% amylose is consumed worldwide. Breeders in BRRI are now concerned with developing high yielding sticky rice simillar to traditional cultivers. With a view to improve low amylose rice for high yield, amylose analysis on 153 local rice cultivars and different germplasm lines were done to help database. Analysis for amylose content showed that there were 75 lines with low amylose (9.0-20.0% and only 7 lines with very low amylose content (9.0%).The line Gijui and C4938-B-B-1-1 had the lowest and the highest amylose content repectively.

Objectives:

To determine amylase content of newly developed low amylose breeding lines for identifying lines with superior grain quality.

Methodology:

The laboratory analysis was conducted at Grain Quality and Nutrition Division of Bangladesh Rice Research Institute, Gazipur. Milled rice out turn was determined by dehulling 200 g rough rice in Satake Rice mill, followed by 75 second polishing in a Satake grain testing mill TM- 05. 153 lines/cultivars which included 34 BRRI/IRRI lines, 31 local cultivars and 88 cultivars from other sources to measure grain amylase content. The samples of these lines/cultivars were milled and amylase content was determined using simplified Amylose Assay of B.O. Juliano.

Achievement/Findings:

The lines/cultivars varied in amylase content from 7.2 to 29.7%. The line Gijui and C4938-B-B-1-1 had the lowest and the highest amylase content respectively. Thirty five lines had high amylase content (>25.0%), 36 lines had intermediate and 75 lines had low amylase (9.0-20.0%). Only 7 lines had very low amylase (2.0-9.0%).
